响应面分析白灵菇菌糠发酵饲料加工调制与优化研究 被引量:2

摘要 为提高白灵菇菌糠发酵饲料中粗蛋白质含量,本试验以假丝酵母菌和嗜酸乳杆菌为发酵液,以发酵后白灵菇菌糠粗蛋白质含量为响应值,通过响应面分析法中Plackett-Burman设计,对影响白灵菇菌糠发酵效果的相关因素进行了分析并成功筛选出主效应因子。在PB设计基础上,根据主效应因子作用大小与方向进行了爬坡试验,再进行Box-Behnken中心复合试验,对主效应因子进一步优化,建立了白灵菇菌糠发酵模型。响应面优化试验结果表明:白灵菇菌糠发酵的最佳工艺条件为白灵菇菌糠∶玉米粉质量为19.1∶1,假丝酵母菌与嗜酸乳杆菌体积比为7.0∶1,菌液接种量7.1%,料水比为2.1∶1,初始p H值为5.0。在此条件下粗蛋白质含量预测值为17.70%,验证试验得到实际粗蛋白质含量为17.41%,与理论预测值相比偏差较小。与发酵前菌糠中粗蛋白质含量相比,提高了78.6%,菌糠中假丝酵母菌和嗜酸乳杆菌最大活菌数为8.7×109cfu/m L,在常温条件下贮藏60 d后,菌糠中的活菌数仍然保持在9.8×107cfu/m L,经检测菌糠中霉菌毒素含量低于国家饲料卫生安全限量标准。 The present research boosted crude protein in pleurotus nebrodensis residues through fermentation with Candida albicans and Lactobacillus acidophilus,and then with crude protein of the fermented residues as the response value, Plackett-Burman design(PBD) and response surface methodology(RSM) were applied to identify the numerous variables influencing the fermentation process and the essential factors were screened.Based on Plackett-Burman design,Box-Behnken Design method was adopted following a slope-climbing test on the direction and magnitude of the main effect factors to optimize these crucial factors and thus a model for pleurotus nebrodensis residues fermentation was eatablished.The optimal levels and interactions of the significant factors found for the fermentation process using RSM was 19.1∶1for the mass ratio of pleurotus nebrodensis residue to corn flour,7.0∶1 for the volume ratio of Candida albicans to Lactobacillus acidophilus,7.1% for the inoculation of bacteria solution,2.1∶1 for the ratio of material to water, and 5.0 for the initial p H value.Under these optimized conditions,the predicted value of crude protein was 17.70% and the observed experimental result was17.41%.The observed and predicated values were regarded to be relatively close.The fermented residues presented an increase of 78.6% in crude protein content, while the maximum viable count of Candida albicans and Lactobacillus acidophilus reached 8.7×109cfu/m L.The viable count stood at 9.8×107cfu/m L after storage at room temperature for 60 days, with mycotoxin content within the national limit of feed safety.
