家蚕茧壳水解物对小鼠细胞免疫功能的影响 被引量:3

摘要 以小鼠为实验动物研究家蚕茧壳水解物对机体细胞免疫功能的影响。分别给正常雄性BALB/c小鼠灌胃低剂量(50mg/kg)、中剂量(200mg/kg)和高剂量(500mg/kg)的家蚕茧壳水解物6周,然后利用MTT法检测小鼠脾淋巴细胞增殖能力;利用血液涂片的方法对小鼠外周血白细胞进行分类计数;利用FACS检测CD4+T细胞和CD8+T细胞比率;利用抗体芯片方法检测细胞因子分泌量。结果显示,中剂量家蚕茧壳水解物可极显著提高小鼠脾淋巴细胞转化率(P<0.01),高剂量组也表现出显著作用(P<0.05),并且和对照相比,中剂量提高的量是对照的4倍;细胞涂片结果显示,摄取家蚕茧壳水解物的小鼠和对照组相比,嗜酸性粒细胞和嗜中性粒细胞的比率下降,而单核细胞和淋巴细胞的比率上升;中剂量组CD4+T细胞和CD8+T细胞比率显著提高(P<0.05);IL-17、IL-6、IFN-γ3种细胞因子的分泌增多,其中,中剂量组IL-17分泌量达对照组的4.5倍。综上所述,家蚕茧壳水解物能够增强小鼠的细胞免疫功能。 To explore the effect of silk cocoon shell hydrolysates on cellular immunity of normal mice,the proliferation of the lymphocytes was detected by MTT assay; the blood leukocytes were classified and counted by using the method of blood smear; the ratio of CD4^+T and CD8^+T was detected by FACS; and the secretion of cytokines was detected by antibody microarrayfollowing the male BALB / c mice were respectively fed with low( 50 mg / kg),middle( 200 mg / kg) and high dose( 500 mg / kg) of silk cocoon shell hydrolysates for 6 weeks. Compared to control group,the spleen lymphocyte proliferation ability of the mice in high( P 〈0. 05) and middle( P 〈0. 01) dose groups were significantly increased. While the value of the middle dose group was 4 times of the control group.. In addition,in the blood leukocytes of the group treated with silk cocoon shell hydrolysates,the ratio of eosinophils and neutrophils significantly decreased and the ratio of mononuclear cells and lymphocytes remarkably increased. The ratio of CD4^+T and CD8^+T in the middle dose group was significantly increased( P 〈0. 05). The silk cocoon shell hydrolysates promoted the mice cytokines secreting such as IL- 17、IL- 6、IFN- γ,and the secretion of IL- 17 of the middle dose group was 4. 5 times of the control. These results show that the silk cocoon shell hydrolysates enhancs the cellular immunity of normal mice.
