摘要
目的探讨NK-1受体(NK-1R)在黏膜接触点性头痛患者接触点鼻黏膜组织中的表达情况。方法采用免疫组织化学方法检测NK-1R在18例黏膜接触点性头痛患者接触点和非接触点鼻黏膜组织中的表达,应用逆转录聚合酶链反应(RT-PCR)技术检测部分NK-1R表达阳性的接触点和相应非接触点鼻黏膜组织中的NK-1R和P物质(SP)的mRNA水平。结果 NK-1R主要表达于接触点鼻黏膜组织的腺泡上皮细胞、炎性细胞和神经纤维。与非接触点鼻黏膜组织相比,接触点鼻黏膜组织中的NK-1R和SP的mRNA水平显著上调。结论 NK-1R在接触点鼻黏膜组织内的表达可促进痛觉信号的传递,可能参与黏膜接触点性头痛的发生,SP可能是其传递疼痛信息的重要介质。
Objective To investigate the expression of neurokinin-1 receptor (NK-1R) in nasal mucosa tissue from pa- tients with intranasal mucosal contact point headaches. Methods The expression of NK-1R was detected by immuno- histochemical staining method in mucosal contact points and non-contact point from 18 patients respectively. The mRNA level of NK-1R and substance P (SP) was detected by real-time quantitative reverse transcription polymerase chain reaction (RT-PCR) at NK-I R positive cells at mucosal contact point and non-contact point respectively. Results NK-I R was mainly located in acinar epithelial cells, inflammatory cells and nerve fiber of contact point nasal mucosa. The mR- NA of SP and NK-I R were up-regulated in mucosal contact point compared with those in mucosal non-contact point. Conclusion NK-1R, as a mediator of pain transmission, may play an important role in the occurrence of intranasal mucosal contact point headaches.
