摘要 目的:观察14周龄自发性高血压大鼠(SHR)体外培养骨髓来源内皮祖细胞(EPCs)数量及功能的变化。方法:随机将自发性高血压大鼠及正常血压WKY大鼠分为2组,每组各12只。以密度梯度离心法分离骨髓来源单个核细胞体外培养,差速贴壁法培养后收集二次贴壁细胞并行鉴定为骨髓来源内皮祖细胞,测定EPCs数量、增殖活力(CCK8法)、迁移、黏附能力和NO分泌功能。结果:14周龄SHR骨髓来源EPC数量与14周龄WKY大鼠比较明显减少(P<0.01),细胞增殖、迁移、黏附能力和NO分泌能力受损(P<0.01)。结论:14周龄SHR大鼠比Wistar大鼠骨髓来源内皮祖细胞数量减少,功能减退。 Objective To investigate the quantity and function of bone marrow - derived endothelial progenitor cells in 14 - week - old spontaneously hypertensive rats. Method 14 -week -old SHR and WKY rats were respectively randomized into two group,with 12 rats ineach group. Mononuclear cells were harvested fromrat bone marrow by density gradient centrifugation. Adherent cells were collected by differential velocity adhcl:ent technique identified as bone marrow - derivedendothelial progenitor cell. To evaulate the quantity, proliferative activity (CCK8 method), migration, proliferation ability, adhesion and NO secreting function ofendothelial progenitor cell. Results Compared with 14 - week - old WKY rats, the quantity of 14 - week - old SHR were significantly reduced ( P 〈 0. 01 ) , the migration ability, prolifera- tion, adhesion and NO" secreting function of 14 - week - old spontaneously hypertensive rats of endothelial progenitor cell were significantly impaired( P 〈0. 01 ). Conclusion Compared with 14 -weekold WKY rats ,the quantity of bone marrow- derived endothelial progenitor cell in 14 -week -old spontaneously hypertensive rat were reduced, function significantly decline.
