苦参素促进EAE大鼠白细胞介素-10和转化生长因子-β1表达的研究

摘要 目的探讨苦参素对大鼠实验性自身免疫性脑脊髓炎(EAE)的防治作用。方法Wistar大鼠通过局部注射豚鼠全脊髓匀浆和完全弗氏佐剂制成的抗原乳剂诱导EAE模型,应用放射免疫分析法和免疫组化法分别检测实验大鼠血清白细胞介素-10(IL-10)和脊髓腰膨大转化生长因子-β1(TGF—β1)的表达,并观察两种细胞因子的改变与EAE症状的关系。结果苦参素可有效改善EAE症状,并促进EAE大鼠血清中IL-10和脊髓腰膨大TGF—β1的表达。结论苦参素对EAE有明显的防治作用,其作用机制可能与上调IL-10和TGF—β1的相关表达。 Objective To investigate the effect of oxymatrine on experimental autoimmune encephalomyelitis (EAE).Methods The EAE models were made in female Wistar rats by having been immuned with spinal cord extracts together with complete Freund's adjuvant.The serum interlenkin-lO (IL-10)was examined by radioimmunity assay,and the expression of transforming growth factor- betal (TGF-β1) in spinal cord was detected by immunohistochemical techniques.The relationship between changes of IL-10 and TGF-β1 and symptoms of EAE were also investigated.Results Oxymatrine could improve symptotns of EAE,and increase expressions of IL-10 and TGF-β1.Conelusion Oxymatrine showed evident prophylactic and therapeutic effects on EAE,which correlated with increased IL-10 and TGF-β1 expression.
