摘要
目的评估miRNA-214介导电针预处理对大鼠心肌缺血-再灌注损伤的保护作用。方法雄性SD大鼠24只随机分为四组(n=6):假手术组(Sham组)、Sham+电针组(sham+EA)、缺血-再灌注组(I/R组)、缺血-再灌注+电针组(I/R+EA组)。Sham+EA组、I/R+EA组在心肌缺血-再灌注损伤实验前3 d,每天给电针预处理。I/R组及I/R+EA组行心肌缺血-再灌注造模。监测Sham组、sham+EA组、I/R组及I/R+EA组大鼠的心功能指标,并测量心肌梗死面积。检测乳酸脱氢酶、肌酸激酶活性,以及心肌细胞中miRNA-214的表达情况。结果与sham组比较,I/R组心功能指标均降低。I/R+EA组与I/R组比较心功能指标均有所提高。I/R+EA组心肌梗死面积显著小于I/R组。而I/R组乳酸脱氢酶、肌酸激酶活性显著升高并高于I/R+EA组(P<0.05或P<0.01)。Sham组、sham+EA组I/R组及I/R+EA组大鼠的miRNA-214的表达分别为1、1.6、1.75和2.82,I/R组miRNA-214的表达高于sham组,电针预处理增加了sham+EA组和I/R+EA组的miRNA-214表达(P<0.01)。结论 miRNA-214表达上调机制参与电针预处理对大鼠心肌缺血-再灌注损伤的心脏保护作用。
Objective To evaluate whether miRNA-214 mechanism was involved in the cardioprotection by electroacupuncture prctreatment of myocardial ischemia/repeffusion injury in rats. Methods 24 male SD rats were divided into 4 groups randomly ( n = 6 ) : control group (sham group), sham + electric acupuncture group (Sham + EA) group, ischemia/repeffusion group (I/R group), ischemia/reperfusion + electric acupuncture group (I/R + EA group). Rats in group sham + EA and L/R + EA received electroaeu- puncture stinmlation 30 rain per day for 3 days before myocardial ischemia/repeffasion injury. Rats in group I/R and I/R + EA re- ceived left anterior descending (LAD) occlusion for 40rain and repeffusion ibr 180 rain. The indices of the cardiac function, myocardi- al irffaret size , LDH and CK activities as well as expression of miRNA-214 were measured. Results All the indices of the cardiac function were not significantly different between the sham and the sham + EA group. Compared with the sham group, all the indices of the eardiae function decreased significantly in I/R group (P 〈 O. 05 ). I/R + EA group showed that all the indices of the cardiac func- tion increased, compared with I/R group. Myocardial inthrct size in I/R + EA group significm,tly decreased, compared with I/R group. The activity of LDH and CK in of I/R group significantly increased, compared with I/R + EA group ( P 〈 0.05 or P 〈 0.01 ). The expression of miRNA-214 in sham, sham + EA, I/R and I/R + EA group were 1, 1. 6, 1.75 and 2. 82, separately. The expres- sion of miRNA-214 in the L/R group was higher than in sham group. Electroacupuncture pretreatment increased miR - 214 expression in sham +EA group and the I/R + EA group. Conclusions Electroacupuncture pretreatment can protect the heart against myocar- dial ischemia/repeffusion injury and can promote the expression of miRNA-214 in rats.
