Application of fluorescence in situ hybridization technique in the department of pathology

摘要 目的研究荧光原位杂交(fluorescence in situ hybridization,FISH)技术检测肿瘤基因的最佳实验效果。方法选取2014年10月—2015年3月的标本100例,对FISH技术操作进行调整和改进,用电磁炉固定功率煮片,将蛋白酶K替换为胃蛋白酶进行消化,用2×SSC、0.1%NP-40溶液代替甲酰胺洗涤液,严格控制时间、温度及PH值,检测100例肿瘤组织标本中目的基因扩增情况。结果在荧光显微镜100×油镜下观察信号,全部标本杂交成功,信号点明显清晰,有利判读。结论实验室改进技术后,检测肿瘤细胞基因操作更加简单、快速、稳定,杂交成功率最高,从而提高诊断率。 Objective Study the best experimental results of tumor gene by fluorescence in situ hybridization(FISH).Methods 100 cases were chosen from October 2014 to March 2015 to join the study.FISH technical operations was used to adjust and improve, with power cook cooker fixed piece,will replace proteinase K digestion of pepsin,with 2 × SSC.0.1% NP-40 used in place of for- mamide washing liquid,strictly control the time,temperature and PH value,detecting amplification of 100 cases of tumor tissue samples of the target gene.Results In fluorescence microscope 100 × oil were observed signal,all specimens hybridization suc- cess,obviously clear signal point,favorable interpretation.Conclusion The improved laboratory techniques to detect gene manipu- lation tumor ceils were-more simple,fast,stable,and highest success rate of hybridization to improve the diagnostic yield.
