摘要 2型糖尿病是一种慢性的代谢性疾病,发病率呈上升趋势,具有较高的致残率和死亡率。目前对其治疗主要是依靠胰岛素与口服药物治疗,但对此疾病尤其是晚期并发症的控制仍然不理想。近年来,黄连中主要活性成分黄连素被发现对糖尿病有一定的改善和治疗的作用。研究表明,其作用靶点较多,通过多种途径发挥调节糖脂代谢的作用。MiRNA是一种短序列的非编码RNA,在哺乳动物中,它们参与了生物体的生长、发育、衰老以及死亡的调控。研究表明miRNA-29a对糖脂代谢的过程具有重要的调控作用。本研究表明,黄连素可以改善T2DM大鼠胰岛素抵抗的状态,减轻肝脏组织炎性反应,而且对肝脏组织中miRNA-29a的表达具有调控作用。黄连素抗糖尿病的部分机制可能是通过miRNA-29a介导来实现的。 Type 2 diabetes is a chronic metabolic disease and its incidence is increasing gradually,which has high mortality and morbidity. The treatment strategies for T2 DM include injecting insulin and oral agents. However,it is not ideal to control it especially its complications. Berberine,the major activity agent in Chinese herb Huanglian,has a function of anti- diabetes in recent years. The data show that it has multiple targets to regulate glucose and lipid metabolism. MiRNA is a short non- coding RNA and it participates in numbers of process of regulating in mammalian such as growth,development,senescence and death. It is reported that miRNA- 29 a plays an important role in glucose and lipid metabolism. In this study,berberine can improve the state of insulin resistance,reduce the inflammatory response in liver tissue. Moreover,it also can regulate miRNA- 29 a expression pattern. Therefore,the partial mechanism of berberine of anti- diabetes may be mediated by miRNA- 29 a.
