摘要
用高效液相色谱法(HPLC)检测奶糖食品中的海藻糖、葡萄糖、蔗糖和麦芽糖。色谱柱为Xbridge-NH2,检测柱温定为35℃,流动相为乙腈∶水=4∶1,按0.1%的量加氨水,检测流速为1.0m L/min,检测器为示差检测器,此条件下,葡萄糖、蔗糖、麦芽糖和海藻糖能完全的分开,其精密度实验RSD分别为1.6%、1.7%、1%、2.1%;其质量浓度在0~4g/100m L范围内的峰面积与质量浓度成良好线性关系,相关系数R2在0.9997~0.9999之间,回收率在98.4%~99.9%之间。使用该方法测定喔喔奶糖中葡萄糖、蔗糖、麦芽糖和海藻糖的质量分数分别为1.0%、23.0%、15.1%和2.64%。
In this paper, we used high-performance liquid chromatography (HPLC) to detect trehalose, glucose, sucrose and maltose in milk candy. The detecting conditions were : chromatographic column Xbridge-NH2, the column temperature 35℃, the mobile phase acetonitrile : water = 4 : 1 with 0.1% of NH4OH, the flow rate 1.0mL/min, differential detector. Under the above condition, glucose, sucrose, maltose, trehalose could be separated, the precision experiment RSD were 1.6%, 1.7%, 1%, 1.7%, respectively; the peak area had a perfect linear relationship with the concentration of glucose, maltose, trehalose which were in the range of 0.1 - 4g/100mL, the correlation coefficient R2 was between 0.9997 and 0.9999, the recovery rate was between 98.4% and 99.9%. This method was applied in the determination of glucose, sucrose, maltose and trehalose in milk candy with the result of 1.0%, 23.0%, 15.1% and2.64%, respectively.
