摘要
目的探讨氯化镉对人肾小管上皮细胞肾损伤分子-1(KIM-1)mRNA表达的影响。方法人近端肾小管上皮细胞(HK-2细胞)用含10%胎牛血清(FBS)的DMEM培养液传代培养。根据培养液氯化镉浓度分组(10μmol/L、20μmol/L、40μmol/L、80μmol/L)。培养24 h后进行形态观察及生存率检测,用RT-PCR方法检测各组肾小管上皮细胞KIM-1 mRNA的表达。结果氯化镉对人肾小管上皮细胞造成损伤,细胞形态破坏,细胞存活率降低。20μmol/L、40μmol/L浓度氯化镉可增加肾小管上皮细胞KIM-1 mRNA的表达。结论氯化镉可引起人肾小管上皮细胞KIM-1 mRNA表达,预示KIM-1可作为镉致肾小管损伤效应标志物之一。
Objective To explore the influence on expression of KIM- 1 mRNA in HK- 2 cells induced by cadmium chloride. Methods Human renal proximal tubular epithelial cell line HK- 2 cells were cultured in DMEM medium supplemented with 10% FBS. Experiments were conducted in FBS- free medium containing various concentrations of cadmium chloride( 10 μmol / L,20 μmol / L,40 μmol / L and 80 μmol / L).Morphological features and survival rates were detected after incubation for 24 hours. RT- PCR had been applied for measuring the expression of KIM- 1 mRNA. Results Cadmium chloride can injure the morphological features and decrease the viability of cells. The expression of KIM- 1mRNA was significantly increased in HK- 2 cells in concentrations of cadmium chloride at 20 μmol / L and 40 μmol / L. Conclusion Cadmium chloride can induce the expression of KIM- 1 in HK- 2 cells,this indicates that KIM- 1 may be used as an effective marker for renal tubular impairment induced by cadmium chloride.
