高效液相色谱-串联质谱法测定啤酒中4种霉菌毒素

Determination of four mycotoxins in beer by high performance liquid chromatography-tandem mass spectrometry

摘要 目的探讨液相色谱-串联质谱法测定啤酒中4种霉菌毒素的可行性。方法样品采用加入乙腈提取,过Mycosep^(TM)226多功能净化柱净化后,采用HPLC/MS/MS电喷雾电离负离子(ESI^-),以多离子反应监测(MRM)模式检测,外标法定量。结果 4种霉菌毒素的线性范围为5.0μg/L^206.0μg/L,线性相关系数在0.9952~0.9998之间,检出限在0.1~0.2μg/kg之间;低中高3个水平的平均加标回收率在80.6%~91.4%之间;相对标准偏差(RSD)均<6.3%。结论结果提示该方法具有操作简单、干扰少、快速、准确可靠等特点,为检测此类毒素提供了一种新的潜在的方法。研究具有潜在的应用价值。 Objective To investigate the feasible for the determination of four mycotoxins in beer samples by using high performance liquid chromatography-tandem mass spectrometric (HPLC-MS/MS). Methods The four mycotoxins in beer samples were extracted by ultrasonic with acetonitrile. After purified by MycosepTM 226 multi-function column, these target analytes were detected by HPLC-MS/MS-ESI-(MRM model) and quantified with external standard method. Results The calibration curves of four mycotoxins were in the range of 5.0 ~ 206.0 t^g/L,with the correlation coefficients(E) in the range of 0.995 2-0.999 8, the limit of detection of this method was in the range of 0.1 ~ 0.2 I^g/kg; the recoveries of three different spiked concentration were in the range of 80.6%~91.4%, and the RSDs were less than 6.3%. Conclusion The new method possess many advantages, such as easy operation, less interference from impurities, fast and accurate , which is a good potential method for the detection of mycotoxins.
