摘要
目的:观察C57BL小鼠用MPTP进行慢性PD造模后行为学和肾上腺的病理改变。方法:将20只雄性老年C57BL/6J小鼠,随机分为造模组和对照组各10只。造模组给予MPTP药5周,对照组给予相同剂量的生理盐水,造模成功后,观察并统计两组行为学评分:爬杆试验、悬挂实验、游泳实验。取肾上腺进行HE、PAS(糖原染色)、高迁移率族蛋白1(HMGB1)染色,并观察对比两组肾上腺的病理学改变。结果:造模组小鼠和对照组比较,行为学评分差异有统计学意义(P<0.05)。造模组小鼠肾上腺:肾上腺组织有损害,炎性细胞浸润,被膜与各带、髓质之间分界基本不清,皮质层变薄,部分呈局灶性坏死,HMGB1在肾上腺上的表达也明显高于对照组。结论:老化、炎症和肾上腺的结构改变可能与PD的发病有一定的相关性。
Objective:To investigate the changes of adrenal gland histology and behavior in Parkinson’s disease models.Methods:20male C57BL/6Jmice were divided into model group and control group randomly.The mice of model group received bake hypodermic injections of MPTP twice a week for5 consecutive weeks.The mice of control group were given the same volume of normal saline.The physical behavior(pole test,traction test,swim test)of two group animals were analyzed after the experiment.The histopathology of adrenal gland was analyzed and the expression of high mobility group protein(HMGB1),HE,PAS were also determined.Results:As compared with the control group,the model group displayed markedly hypoactive behavior.Significant statistical difference was found between the two groups in three tests(P〈0.05).Histological examination demonstrated severe injuries of the adrenal gland,inflammatory response was apparent,cortices changes thinner and with unclear boundaries,necrosis can been seen.Immunohistochemical staining showed that the density of HMGB1 stain was increased significantly after treatment with MPTP.Conclusion:Parkinson’s disease may be associated with inflammation,aging and the changing of adrenal gland histopathology.
