摘要
目的将人脐血间充质干细胞移植到兔骨折模型中,观察异种移植后的安全性,为人脐血间充质干细胞异种移植提供实验依据。方法制作新西兰兔胫骨横断骨折模型,随机分为细胞移植组、生理盐水组。于骨折模型建立后第3天,在C形臂X线机辅助下垂直于骨折断端进针,分别注射浓度为1×107个/ml经Brd U标记的人脐血间充质干细胞悬液1 ml和等量生理盐水。移植后每天观察一般情况,分别于细胞移植后第3、7天行血常规、血生化检测,采用空气栓塞处死新西兰兔,进行骨折断端组织、脾脏组织病理学观察及免疫荧光染色观察。结果细胞移植后两组一般情况观察无差异;细胞移植后第3天,两组淋巴细胞比率、单核细胞比率比较差异有统计学意义(P<0.05),两组血清生化指标比较差异无统计学意义(P>0.05);移植后第7天,两组血常规、血清生化指标比较差异无统计学意义(P>0.05);细胞移植后第3、7天,细胞移植组骨折断端组织、脾脏组织病理学观察与生理盐水组比较无统计学差异(P>0.05);细胞移植后第7天,细胞移植组在荧光显微镜下均可见散在分布着呈现绿色荧光的阳性细胞。结论异种移植后人脐血间充质干细胞在骨折断端可以大量存活,且未导致明显的免疫排斥反应,因此是安全可行的。
Objective To study the safety of transplantation of human umbilical-cord mesenchymal stem cells( HUMSCs) in local fracture of New Zealand rabbits and approve experimental evidence for the application of HUMSCs. Methods Fracture models were made and were randomly divided into cell transplantation group and saline group. HUMSCs of Brd U-labeled at a concentration of1 × 107 cells / ml and saline were injected into the local fractures by the assistane of C-arm X-ray machine. The rabbits were observed every day for their general reactions before they were sacrificed. Blood samples were collected for hematological and biochemical studies on the 3rd and the 7th day after transplantation. Fracture fragments and spleen organs were inspected pathologically and Immunofluorescence staining was done after air embolism. Results No difference in general reactions after transplantation was observed between the two groups. On the 3rd day after transplantation,there was statistically significant differences in the ratios of lymphocytes and monocytes between the two groups( P〈0. 05),and there was no difference in serum biochemical parameters( P〉0. 05). On the 7th day after transplantation,there was no significant differences in blood routine and serum biochemical parameters( P〉0. 05). There were no significant differences in fracture fragments and spleen organs on the 3rd day and the 7th day between the two groups( P〉0. 05). Scattered positive cells showing green fluorescence were observed under fluorescence microscopy on the 7th day after transplantation. Conclusion Conclusion Heterogenous umbilical cord blood mesenchymal cells can survive a lot and transplantation of heterogenous HUMSCs is safe and feasible without side effects.
