摘要
目的研究人类巨细胞病毒(HCMV)和Epstein-Barr病毒(EBV)及其诱导的肿瘤坏死因子-α(TNF-α)、γ-干扰素(γ-IFN)、白介素(IL)-1β、IL-6、IL-10和IL-12在儿童乳牙根尖周炎中的表达情况。方法收集15例有严重根尖病变的乳牙,取其根尖周肉芽组织作为实验组;同时收集15例无病变的乳牙,取其正常牙髓组织作为对照组。设计并合成引物,通过聚合酶链反应(PCR)检测HCMV、EBV及细胞因子的表达。结果 HCMV(x^2=17.368,P<0.05)和EBV(x^2=15.000,P<0.05)的表达在实验组和对照组之间具有显著性差异;细胞因子包括TNF-α(U=15.372,P<0.05)、γ-IFN(U=16.014,P<0.05)、IL-1β(U=13.833,P<0.05)、IL-6(U=10.416,P<0.05)、IL-10(U=12.105,P<0.05)和IL-12(U=11.961,P<0.05)的表达在两组之间也存在着显著性差异;在根尖周肉芽组织样本中,疱疹病毒(HCMV、EBV)与TNF-α、γ-IFN、IL-1β和IL-12的表达均呈显著正相关(r值为0.289~0.664,均P<0.05)。结论 HCMV和EBV及其诱导的组织破坏性细胞因子的释放可能在儿童乳牙根尖周炎的发病机理中起重要作用。
Objective To study the expressions of human cytomegalovirus (HCMV) and Epstein-Barr virus (EBV) and the expressions of tumor necrosis factor α(TNF-α), y-interferon (y-IIFN) , interleukin (IL)-1β, IL-6, IL-10 and IL-12 induced by them in children's deciduous teeth with periapical periodontitis. Methods A total of 15 deciduous teeth with severe root apex disease were collected, and periapical granulation tissues were obtained to serve as experimental group. Meanwhile, a total of 15 normal deciduous teeth were collected, and pulp tissues were gained to serve as control group. Primers were designed and synthesized, and polymerase chain reaction (PCR) was conducted to detect the expressions of HCMV, EBV and cytokines. Results There were significant differences in the expressions of HCMV (X2 = 17. 368, P 〈 0.05 ) and EBV (X2 = 15. 000,P 〈 0.05 ) between experimental group and control group, and the differences in expressions of cytokines including TNF-α, y-IFN, IL-lβ, IL-6, IL-10 and IL-12 were also significant between two groups ( U value was 15. 372, 16. 014, 13. 833, 10. 416, 12. 105 and 11. 961 , respectively, all P 〈 0.05). In periapical granulation tissue samples, there was a remarkable positive correlation between herpesvirus (HCMV and EBV) and the expressions of TNF-α,y-IFN, IL-1β and IL-12 (r value ranged 0. 289 -0.664, all P 〈 0.05 ). Conclusion HCMV and EBV as well as the tissue-destructive cytokines induced by them may play an important role in the pathogenesis of children' s deciduous teeth with periapical periodontitis.
