利用SSR分析湖南桃江病圃丽江新团黑谷上稻瘟病菌的遗传多样性 被引量:8

Analysis of Genetic Diversity of Magnaporthe oryzae Isolated from LTH by SSR Markers in Rice Blast Nursery at Taojiang in Hunan

摘要 利用抗瘟水稻品种是防治稻瘟病最有效措施,了解稻瘟病菌遗传多样性是抗病品种合理布局的重要基础。本研究于2014年从湖南桃江水稻抗稻瘟病鉴定中心病圃中广谱高感品种丽江新团黑谷(LTH)上分离了120个稻瘟病菌单孢菌株,利用14对SSR引物对其进行遗传多样性分析。结果表明,在相似系数0.75时,120个菌株可分成32个遗传宗谱,其中L1和L7为优势宗谱,分别含有20和16个菌株,各占总菌株数的16.67%和13.33%;宗谱L03,L04,L02分别含菌株11,11,13个,分别占总菌株数的9.17%,9.17%和10.83%;另27个宗谱分别含菌株1~6个,共占总菌株数的40.83%。对比本课题组2012、2013年对湖南稻瘟病菌群体遗传结构的研究结果,在相同的SSR引物、聚类方法以及相似系数下,其湖南桃江病圃中丽江新团黑谷上的稻瘟病菌遗传宗谱数是有所增加的,表明了该病圃中的稻瘟病菌群体复杂的遗传多样性。 Utilization of resistant cultivars is the most effective method to control rice blast,and it is an important basis of rational distribution of resistant varieties to clarify the genetic diversity of M. oryzae. The genetic diversity of 120 isolates of M. oryzae from a broad-spectrum highly susceptibe variety,Lijiangxintuanheigu( LTH) in rice blast nursery at Taojiang,Hunan in 2014,was analyzed by 14 pairs based on SSR markers. The results showed that these isolates were classified into 32 genetic lineages at the 0. 75 similar level,among which L01 and L07 were the dominant lineages,including 20 and 16 isolates,which accounted for 16. 67 % and 13. 33 % in total isolates,respectively;L03,L04 and L02 with 11,11 and 13 isolates separately held 9. 17 %,9. 17 % and 10. 83 % of the total isolates; And the other 27 lineages contained either 1- 6 isolates accounting for 40. 83 % of the total isolates. Contrasting with the results of the genetic structure of M. oryaze isolated from LTH of Hunan by our project group in 2012 and 2013,under the same SSR primers,clustering method and similar level,the genetic lineages of M. oryaze isolated from LTH in 2014 were more than that in 2012,2013. Based on the results,the genetic diversity of M.oryaze in this nursery performed more complex genetic diversity as year increasing.
