摘要
目的探讨白塞病(Behet's disease,BD)患者氧化应激、转录因子NF-E2相关因子(NF-E2-related factor2,Nrf2)和HO-1的表达情况及Nrf2,HO-1与BD的相关性。方法将38例BD患者分为活动期BD组和非活动期(稳定期)BD组,正常组以20例健康人为对照。分别对BD两组患者进行病情评分,采用常规法检测血中ESR和CRP水平、ELISA法检测Nrf2,HO-1和ROS的表达以及化学比色法检测MDA,T-AOC,SOD和GSH的含量。结果活动期BD组病情评分显著高于非活动期BD组(P<0.01);活动期BD组ESR和CRP水平均较非活动期BD和正常组明显升高(P均<0.01)。与正常组比较,BD两组血清ROS和MDA水平均明显升高(P<0.05),T-AOC,SOD和GSH水平均显著下降(P<0.05);而活动期BD组较非活动期BD组表达ROS,MDA,T-AOC,SOD和GSH差异明显(P<0.05)。活动期BD组Nrf2和HO-1水平较非活动期BD组和正常组下降(P<0.05),与病情评分、ESR和CRP呈负相关(P<0.05),尤以Nrf2与病情评分和CRP相关性强(P<0.01)。结论 BD存在氧化应激异常;在活动期Nrf2和HO-1下降,并随着病情加重而降低;Nrf2的下降可能与BD的发展和加重密切相关。
Objective To explore the expression of oxidative stress, NF-E2 related factor (Nrf2) and HO-1 in patients with Beheet's disease (BD) and the correlation between Nrf2/HO-1 and BD. Methods A total of 38 patients with BD were divided into active BD group and inactive BD group, while 20 healthy individuals as control. Severity scores of patients in two BD groups were respectively evaluated. Meanwhile, routine test was applied for the detection of the levels of ERS and CRP, ELISA method for testing the expression of ROS, Nrf2 and HO-1, and chemical colorimetric method for detecting the content of MDA, T-AOC, SOD and GSH. Results Severity scores of active BD patients were considerably higher than those of inactive BD patients (P 〈0. 01 ). At the same time, ESR and CRP levels in active BD group remarkably ascended in comparison with those in inactive BD group and normal group (P 〈 0. 01 ). Compared to normal group, serum levels of ROS and MDA in two BD groups significantly increased ( P 〈 0. 05 ) , while T-AOC, SOD and GSH levels were decreased (P 〈 0. 05). The significant differences of expression of ROS, MDA, T-AOC, SOD and GSH were observed between active BD group and inactive BD group. On the other hand, the levels of Nrf2 and HO-1 in active BD group were lower than those in inactive BD group and normal group (P 〈 0. 05) , which was negatively correlated with severity score, ESR and CRP ( P 〈 0. 05 ), especially Nrf2 showing a strong correlation with severity score and CRP (P 〈 0. 01 ). Conclusion There is an abnormal oxidative stress indeed existing in BD. Moreover, a reduction of Nrf2 and HO-1 expression occurs in active BD, which gradually decreases with the aggravation of the disease. Thus, the decline of Nrf2 may be closely related with the development and exacerbation of BD.
