摘要
目的观察外源性硫化氢(Hydrogen Sulfide,H2S)对血管性痴呆大鼠(vascular dementia,Va D)海马NF-κB及环氧合酶-2表达的影响。方法采用改良的四血管法(4-VO)复制大鼠Va D模型,将模型大鼠按给药干预时间分为1 d、7 d、30 d 3个时段,每时段又分为假手术组、模型组、低剂量组、高剂量组。低剂量组每日腹腔注射H2S供体Na HS 30μmol/kg,高剂量组每日腹腔注射H2S供体Na HS 100μmol/kg,假手术组与模型组每日腹腔注射等量生理盐水,然后进行Morris水迷宫检测大鼠学习记忆能力,Western blot法检测大鼠海马区NF-κB、COX-2的表达。结果与假手术组比,3个时段的模型组大鼠学习记忆力均明显下降,NF-κB、COX-2的表达显著提高;与模型组相比,硫氢化钠干预组可改善大鼠的学习记忆,且低剂量组COX-2、NF-κB的表达明显降低(P<0.05)。结论硫氢化钠可改善血管性痴呆大鼠的认知水平,可能是通过抑制大鼠脑组织NF-κB、COX-2的表达发挥其脑保护作用。
Objective To observe the effcets of exogenous hydrogen sulfideand ( H2S) on he expression of nuclear factor-Kappa B(NF-κB) and cyclooxygenase-2(COX-2) in the hippocampus of vascular dementia(VaD) rats. Methods A modified four vessels occlusion method (4-VO) was used for VaD model rats preparation,the rats were divided into three periods by drug intervention time : 1 day period, 7 days period, 30 days period. Every period were divided into four subgroups:sham-operated group, model group, low-dose group, high-dose group. The rats of low-dose group and the high-dose group were intraperitoneal injected with Naris (H2S donor) 30 μmo/kg and 100 μmol/kg per day ,and the sham-operated group and model group were intraperitoneal injected with saline. Then the Morris water maze test was used to detect the learning and memory abilities of the rats. The expression of COX-2 and NF-KB were measured by Western blot. Results Compared with sham-operated group,the model group rats had greatly reduced the Learning memory. Expression of NF-κB and COX-2 in model groups were higher than sham-operation groups ( P 〈 0.05 ). Compared with model group, the rats injected NariS promoted the learning memory and expression of COX-2 and NF-KB in low-dose group rats were significantly lower expressed (P 〈 0.05). Conclusion NariS may improve cognitive level in VaD rats, and its protective effect on brain may be probably via inhibiting expression of NF-κB and COX-2 in the brain.
