摘要
[目的]优化蒙山牛肝菌液体发酵培养基。[方法]以菌丝干重为指标,首先采用单因素试验筛选适于菌丝体生长的最佳碳氮源,再以正交试验优化碳源与氮源以及无机盐与VB1的最佳配比。[结果]蒙山野生牛肝菌优化的液体培养基配方为乳糖10 g/L,可溶性淀粉20 g/L,酵母膏5 g/L,牛肉膏10 g/L,KH2PO41 g/L,Mg SO4·7H_2O 1 g/L。[结论]正交设计法优化的蒙山牛肝菌液体发酵培养基具有一定的应用价值,可为大规模发酵生产及后继研究提供基础数据。
[ Objective ] The aim was to optimize liquid fermentation medium of Mengshan mountain boletus. [ Method ] Adopting mycelial dry of Mengshan mountain boletus as major index, firstly the single factor method was used to select the best carbon source and nitrogen source for mycelium growth, and then the optimum ratio of carbon source, nitrogen source, inorganic salt and VBt was optimized by the orthogonal experiment. [Result] The optional liquid fermentation medium of Mengshan mountain boletus were as follow: lactose of 10 g/L,soluble starch of 20 g/L, yeast extract of 5 g/L, beef paste of 10 g/L, KH2PO4 Of 1 g/L, MgSO4 ·7H2O of 1 g/L. [ Conclusion] The optimized liquid fermentation culture medium of Mengshan mountain boletus by the orthogonal design has a certain application value, and can provide the basic data for large-scale fermentation and subsequent research.
