摘要
目的研究叉状头转录因子p3(Foxp3)在呼吸道合胞病毒(RSV)至毛细支气管炎模型大鼠中的表达情况及其意义。方法将30只大鼠完全随机分为对照组和RSV组,每组15只,RSV组大鼠采用滴鼻法建立RSV毛细支气管炎模型。观察2组大鼠肺组织病理变化;比较2组大鼠外周血单个核细胞(PBMC)中和肺组织中Foxp3 mRNA水平及肺组织中Foxp3蛋白表达水平差异;比较2组大鼠血浆白细胞介素10(IL-10)和转化生长因子β(TGF-β)水平差异。结果RSV组大鼠肺组织肺间质炎症严重,肺泡间隔增宽,伴大量淋巴细胞浸润,对照组肺组织无异常表现;RSV组PBMC和肺组织中Foxp3 mRNA水平均明显低于对照组[相对值:(0.15±0.03)比(1.00±0.00),(0.26±0.05)比(1.00±0.00)],差异有统计学意义(P〈0.01);RSV组肺组织中Foxp3蛋白水平明显低于对照组;RSV组大鼠血浆IL-10和TGF-β表达水平明显低于对照组[(86±7) μg/L比(118±8) μg/L,(22±4) μg/L比(43±5) μg/L],差异有统计学意义(P〈0.05)。结论Foxp3在RSV毛细支气管炎中表达下调,可能是导致毛细支气管炎发病的重要机制。
ObjectiveTo study the expression and significance of forkhead box p3 (Foxp3) in respiratory syncytial virus (RSV) induced capillary bronchitis. MethodsThirty rats were randomly divided into control group and RSV group (n=15 in each group). The capillary bronchitis was induced by RSV in RSV group. The pathological changes of lung tissue was observed; the mRNA levels of Foxp3 in peripheral blood mononuclear cell (PBMC) and lung tissue, the expression of Foxp3 in lung tissue, the levels of interleukin-10 (IL-10) and transforming growth factor-β (TGF-β) in plasma were compared between groups. ResultsThe rats in RSV group showed severe inflammation in pulmonary interstitial, with wider alveolar septa and infiltration of a large number of lymphocyte; the rats in control group had no abnormal performance. The mRNA levels of Foxp3 in PBMC and lung tissue, the expression of Foxp3 in lung tissue were significantly lower in RSV group compared with those in control group [(0.15±0.03) vs (1.00±0.00),(0.26±0.05) vs (1.00±0.00)](P〈0.05); the levels of IL-10 and TGF-β in plasma were significantly lower in RSV group compared with those in control group [(86±7) μg/L vs (118±8) μg/L, (22±4) μg/L vs (43±5) μg/L] (P〈0.05). ConclusionThe down regulation of Foxp3 could be an important mechanism of RSV induced capillary bronchitis.
