摘要 为了研究离子色谱法测定烟叶中游离氨基酸含量,以游离氨基酸测定总量为指标,考察了萃取所用的溶剂、温度、时间以及料液比等因素对游离氨基酸测定量的影响。结果表明,烟草中游离氨基酸的最优检测条件为:萃取溶剂为0.1 mol/L盐酸、萃取时间为30 min、萃取温度为30℃、烟叶与盐酸的比例为1∶40(w/v)。测定方法的氨基酸检出限为2.76~22.36 ng/m L,定量限为9.57~102.13 ng/m L,试验的RSD为1.26%~4.96%,氨基酸的加标回收率为83.51%~106.78%,说明该优化方法可行,利用确定的最佳条件测定了国内主产区的10种烤烟型烟叶。 In order to study the effect of ion chromatography method for the determination of free amino acids in tobacco,the optimization of the experimental conditions were studied, the value of free amino acids used as a response value. The experimental results showed that the best experimental conditions were as follows : the extract solvent was 0.1 mol / L hydrochloric acid; the temperature of experiment was 30 ℃; the extract time was 30 min; the solid to liquid ratio was 1∶40(w / v). The detection limit of the method was 2.76~22.36 ng / m L,limit of quantitation was 9.57~102.13 ng / m L, the method has high recovery(83.51%~106.78%)and good repeatability(RSD5%), these results demonstrate that this method is feasible. The contents of free amino acids in 10 flue-cured tobaccos were determined.
