百草枯诱导大鼠肺纤维化的机制分析 被引量:9

摘要 目的观察百草枯(paraquat,PQ)所致Wistar大鼠肺纤维化的病理改变,探讨可能的机制。方法将Wistar大鼠随机分为对照组、AG490组、PQ模型组、AG490+PQ组。PQ组按一次性给予50 mg/kg PQ灌胃染毒;AG490组给予8 mg/kg AG490皮下注射;AG490+PQ组造模前0.5 h给予8 mg/kg AG490皮下注射,再给予50 mg/kg PQ灌胃染毒;对照组给予1 ml蒸馏水灌胃。造模后第7天、14天、28天分别处死动物,第7天、14天、28天测定肺泡灌洗液中细胞总数和计算PMN百分比,苏木精-伊红(HE)染色观察肺泡炎症,第28天测定肺组织中TGF-β1m RNA表达水平,Masson染色观察肺纤维化程度。结果第7天、14天和28天PQ组大鼠较对照组的肺泡灌洗液中白细胞数量明显增加,PMN数量也显著增多(P<0.05),肺泡内有炎症细胞侵润,肺结构破坏;第28天,Masson染色显示胶原纤维沉积,肺纤维化形成,PQ组大鼠肺组织TGF-β1m RNA表达水平较对照组明显升高(P<0.05),AG490+PQ组TGF-β1m RNA表达水平较PQ组显著减少(P<0.05)。结论 TGF-β1参与PQ中毒所致肺纤维化形成,其机制可能与STAT信号转导通路有关。 Objective To observe paraquat-induced pulmonary fibrosis in Wistar mice pathologic change and research on its mechanism. Methods The Wistar mice were randomly divided into 4 groups: Control group, AG490 group, PQ model group and AG490 + PQ group. The PQ group were administrated paraquat stomach in 50 mg/kg, ACA90 group received an subcutaneous injection of 8 mg/kg,ACA90+PQ group received an subcutaneous injection of AG490 before 0.5 hour in mondling, control group were administrated paraquat stomach in H2O of 1 ml,the Wistar mice were sacrificed at the 7 th, 14 th and 28 th day. The degree of lung inflammation were assessed by total cell and classification of PMN in bronchoalveolar lavage fluid and Hematoxylin-eosin ( HE ) staining at the 7 th day, the degree of pulmonary fibrosis were observed by TGF-β1 mRNA expression and Masson dyeing at the 28 th day. Results The PQ group show the increase total cell and classification of PMN in bronchoalveolar lavage fluid as well as increase aveolar inflammation compared with Control group at the 7 th, 14 th and 28 th day(P〈0.05) ;The more collagen deposition , TGF -β1 mRNA expression in PQ group compared with control group at the 28 th(P〈0.05) , The less collagen deposition, TGF-β1 mRNA expression in AG490 +PQ group compared with PQ group control group. Conclusion The TGF-β1 Participate the paraquat-induced pulmonary fibrosis in Wistar mice ,The probable mechanism might be associated with the STAT signal transduction pathway.
