摘要
探索磷脂酶C酶活适宜条件,及其对菜籽油脱胶的优化方案。用卵黄琼脂平板法和NPPC法对磷脂酶C的酶活条件进行检测;在单因素试验的基础上,利用Box-Behnken试验设计和响应面分析法探讨了反应温度、pH、时间和加酶量对菜籽油脱胶效果的影响。结果:确定磷脂酶C最佳反应温度和pH分别为57℃和7.0;酶法脱胶最佳优化方案为:温度60℃,加酶量0.05%,pH 6.9,反应时间3.5 h。在此方案下得出最终磷含量为9.6 mg/kg,脱胶率达94.75%。
The suitable conditions of phospholipase C enzyme activity and the optimization solution of rapeseed oil degumming was explored. The phospholipase C enzymatic activity conditions were detected by using egg yolk agar plate method and NPPC method; On the basis of single factor experiments, reaction temperature, pH, time and enzyme concentration were optimized by the Box-Behnken design and response surface. The optimum reaction temperature and pH of phospholipase C were 57 ℃ and 7.0 respectively; The best optimization scheme for enzymatic degumming was as followings: temperature 60 ℃, enzyme amount 0.05%, pH 6.9, the reaction time of 3.5 h. Under this scheme the final phosphorus content arrived at 9.6 mg/kg, and the degumming rate was 94.75%.
