番茄SlHB8基因的克隆及其对非生物胁迫的响应

Cloning of SlHB8 Gene From Tomato and Its Response to Abiotic Stress

摘要 HD-Zip Ⅲ转录因子家族在植物的生长发育中发挥着重要作用。以Micro-Tom番茄为试材,采用RT-PCR、荧光定量PCR以及生物信息学的方法,研究了番茄SlHB8基因的序列特征、表达模式以及在非生物胁迫下的响应,以期为鉴定SlHB8基因在番茄中的功能奠定基础。结果表明:番茄中含有1个编码HD-Zip Ⅲ转录因子的基因SlHB8。该基因全长2 523 bp,编码841个氨基酸,含有HD、bZip、START和MEKHLA 4个保守结构域,与拟南芥的ATHB8同源性最高。SlHB8在番茄各组织中均有表达,在茎中的表达量最高。高低温、干旱、伤害、乙烯、水杨酸处理均能诱导该基因下调表达,而赤霉素、脱落酸、生长素处理均能使该基因的表达先增加再下降。利用PLACE和PlantCARE对SlHB8启动子区域的顺式作用元件进行预测分析发现,SlHB8启动子含有赤霉素、脱落酸、水杨酸、热激、伤害、干旱等顺式作用元件。表明SlHB8可能在番茄的非生物胁迫应答中发挥重要调控作用。 The HD-ZipⅢtranscription factor family plays an important role in plant growth and development.In this study,Micro-Tom tomato was used as material.Using RT-PCR,fluorescence quantitative PCR and bioinformatics methods,the SlHB8 gene′s sequence characteristics,expression patterns and responses under abiotic stress were studied so as to lay a foundation for identifying the function of SlHB8 gene in tomatoes.The results showed that there was one gene encoding the HD-ZipⅢtranscription factor(SlHB8)in tomato.The full length of SlHB8 was 2 523 bp,encoding 841 amino acids with four conserved domains of HD,bZip,START and MEKHLA,showing the highest homology with ATHB8 of Arabidopsis thaliana.SlHB8 was expressed in all tomato tissues,and the highest expression was found in stem.All these treatments,such as cold,heat,drought,wound,ethylene(ETH),and salicylic acid(SA)could reduce the expression level of this gene.Gibberellin(GA),abscisic acid(ABA),and auxin treatment could make the expression level of the gene to increase first and then decrease.Promoter sequence analysis by PLACE and PlantCARE revealed that there were many cis-acting elements such as gibberellin(GA),abscisic acid(ABA),salicylic acid(SA),heat shock,wound,and drought.These results indicated that SlHB8 might play an important role in regulating the abiotic stress response of tomato.
