汽车尾气染毒动物模型的构建及其对雄鼠生殖功能影响的初步研究 被引量:2

摘要 目的探讨汽车尾气染毒动物模型的构建方法,并初步研究其对雄性C57BL/6小鼠生殖系统的影响。方法设计构建汽车尾气染毒装置,观测该装置内氧气含量的变化和小鼠生存状态,明确通入汽车尾气的时间和间隔;为了评估本染毒装置的可靠性,将24只C57BL/6小鼠随机分成染毒组和对照组,染毒组予以汽车尾气暴露,而对照组予以正常空气。40 d后对比2组生殖腺质量的变化、精子活力、精子数目和精子畸形率,并检测血和睾丸中活性氧含量(ROS)。结果向染毒装置持续4 min通入汽车尾气,此时装置内氧气含量为(17.33±0.09)%,间隔20 min后装置内氧气的含量接近空气中的含量。染毒模型构建结束后,染毒组小鼠的生殖腺质量、精液质量明显下降,而血和睾丸ROS的含量均高于对照组。结论该研究为构建汽车尾气的染毒模型提供了方法且效果良好,明确了汽车尾气染毒后可损伤生殖系统。 Objective The purpose of this research was to discuss the method of the automobile exhaust exposed animal model and to determine the effects of automobile exhaust exposure on male C57BL/6 mouse reproductive system. Methods An automobile exhaust ex- posed device was designed and made. By detecting the change of the oxygen content of the device and C57BL/6 mice could be alive or not, we made sure the time and time interval of vehicle exhaust emissions. In order to assess the reliability of the automobile exhaust exposed de- vice ,24 male C57BL/6 mice were divided into two groups randomly. Automobile exhaust exposure group lived with high concentration of auto- mobile exhaust for 40 days, control group was suffered from fresh air. The effect of automobile exhaust on the mouse reproductive system was measured by comparing the two group changes of sexual gland weight and semen quality ( sperm motility, sperm abnormol rate and sperm count) and evaluated the oxidative stress by detecting ROS( serum and testicular tissue). Results We made the 4 rain as the time of vehi- cle exhaust emissions and oxygen content was (17.33 ± 0.09)% in the device. Within 20 min oxygen content in the device was close to the content of oxygen in the air. After automobile exhaust exposure, we found that the average gonad weight and sperm semen quality were de- clined significantly in the automobile exhaust exposure group compared to the control group (P 〈 0.05 ) and ROS in automobile exhaust expo- sure group were higher than it in the control group ( P 〈 0.05 ). Conclusion Our work provides a good model of automobile exhaust expo- sure and makes sure that automobile exhaust exposure is harmful to male reproductive system.
