摘要
目的本文主要研究近几年来NCBI数据库最新释放出来的8种新型HPV即HPV(126,135,136,137,140,144,154,166),对其进行分子进化树分析,进行分类并设计特异型引物。方法 NCBI检索8种新型HPV的基因组序列,然后用Blast相似性比较得到与其L1高度相似的HPV,其次选择HPVαβγ各属具代表性的HPV,最后用Clustal W对以上所有58种HPV L1区氨基酸序列进行比较分析,构建分子进化树;进一步用Primer-Blast对8种新型HPV进行特异型引物设计。结果系统进化树分析得出8种HPV均属于γ属,利用Primer-Blast分析得到这8种HPV特异型引物。结论 HPV126、HPV135、HPV136、HPV137、HPV140、HPV144、HPV154、HPV166均为γ属。
Objective This study was based on the latest reports of 8 novel types of HPV (human papillomavirus) genomes inNCBI. The eight HPVs were H PV (126, 135, 136, 137, 140, 144, 154 and 166). The phylogenetic tree was constructed,the boxshadow was built based on the L1 amino acid sequence and the specific primers were designed based on the L1 nucleotidesequence. Methods The similar HPVs to the 8 novel types of HPVs was searched by Blast, and the representative HPVs ofa, 3, 7 genus were selected, and all of the 58 HPV L1 amino acid sequences were analyzed and compared by the Clustal W soft-ware to construct the phylogenetic tree. The specific primers were designed by the primer - blast. Results The phylogenetictree suggested that all the eight HPVs were from 7 genu. Using the primer - blast, we designed the specific primers of eachHPV. Conclusions The HPV types of 126, 135, 136, 137, 140, 144, 154 and 166 are all in 7 genus.
