Analysis of allele sharing in genetic relationship

摘要 目的探讨常见亲缘关系中等位基因共享的特点。方法利用自主研发的软件,在GoldeneyeTM20A系统中模拟100 000组无关个体、父子、同胞、半同胞、一代/二代堂表兄弟,对所得数据进行统计分析,计算各组的等位基因共享类型和数量分布特征。结果等位基因共享与亲缘关系之间存在显著相关性。共享类型分布特点:随着亲缘关系的疏远,等位基因完全共享比例逐渐降低,分布类型逐渐趋近无关个体。共享数量分布特点:随着亲缘关系的疏远,等位基因共享数量的均数逐渐降低。其中同胞组均数最高,无关个体组均数最低。同胞和半同胞与无关个体的分布差异较显著,而一代/二代堂表兄弟与无关个体分布差异较小。结论等位基因共享特点有助于分析同胞和半同胞关系。 Objective To explore the characteristic of allele sharing in genetic relationship. Methods For the typing data of 19 autosomal STR loci with the GoldeneyeTM 20A, the characteristic of allele sharing between unrelated-individual, parent-child, full-sibling, half-sibling, first-cousin and second-cousin pairs from one hundred thousand cases of computer simulating genetic relationship testing were analyzed respectively, and the type and number distribution of alleles sharing in each group were calculated. Results A significant correlation between the type and number of shared alleles and genetic relationship was observed. The type distribution characteristics of shared alleles showed that the more distant genetic relationship, the lower the proportion of allele sharing, and the distribution of allele sharing gradually approached unrelated individuals group. Similarly, the number distribution characteristics of shared alleles showed that the more distant genetic relationship, the lower the mean of allele sharing number. The mean of full-sibling group was the highest, and unrelated-individual group was the lowest. The distribution difference between fulL/half-sibling group and unrelated-individual group was relatively significant, while the distribution difference between first/second-cousin group and unrelated-individual group was relatively insignificant. Conclusion The characteristic of allele sharing is helpful for analyzing the full-sibling and half-sibling relationship.
