益智健脑颗粒对老年大鼠空间学习记忆及中枢胆碱能系统的影响 被引量:2

The effect of yizhi jiannao granule on the abilities of learning and memory and cholinergic system in aged rats

摘要 目的:观察益智健脑颗粒对老年大鼠空间学习记忆及中枢胆碱能系统的影响。方法:以自然衰老大鼠为研究对象,通过Morris水迷宫实验、尼氏染色及免疫组织化学等方法的检测,观察益智健脑颗粒对老年大鼠空间学习记忆、基底前脑胆碱能神经元及海马CA1与CA3区和第一躯体感觉区(S1Tr)M1受体阳性神经元的影响。结果:与老年对照组相比,益智健脑颗粒(10g/kg)治疗组平均逃避潜伏期至第四天两者有极显著差异,其空间探索实验中穿越平台次数及潜伏期呈极显著性差异;在内侧隔核(MSN)与斜角带核(VDB)内,胆碱能神经元数目增多,两者有显著性差异,在Meynert核(NBM)内有极显著性差异;海马区与大脑皮质神经元、M1受体阳性神经元数目有所增加,且在CA1与CA3区和S1Tr内有极显著性差异。结论:益智健脑颗粒能改善自然衰老大鼠空间学习记忆能力,机制可能是通过增强基底前脑胆碱能神经元Ch AT的表达水平,提高乙酰胆碱合成量,海马及大脑皮质胆碱能毒蕈碱M1受体表达水平上调则可接受更多来自基底前脑的投射,触发受体后效应,进而对衰老大鼠中枢胆碱能系统具有一定的保护作用。 Objective: To observe the effect of yizhi jiannao granule on the abilities of learning and memory and cholinergic system in aged rats.Method: To observe the influence of yizhi jiannao granule on learning and memory in rats aged,basal forebrain cholinergic neurons and the M1 receptor positive neurons of hippocampal CA1 and CA3 area and S1 Tr,natural aging rats were detected by Morris water maze test,Nissl staining and immunohistochemistry methods. Result: The average escape latency of yizhi jiannao granule( 10 g / kg) treated group was significantly shorter than the aged control group since the fourth day,and there was a significant difference( P〈0. 01). Space exploration experiments showed that there was a significant difference between rats in older group and yizhi jiannao granule( 10 g / kg) treated group in the number of crossing the platform( P〈0. 01). The Ch AT-positive cells of the rats in aged treatment group were stained deeply and their neuronal processes are more obvious. Compared with the rats in aged control group,Ch AT positive cell count in the basal forebrain MSN and VDB of the rats in yizhi jiannao granule( 10 g / kg) treated group was increased,and there are significant differences between them( P〈0. 05); in NBM there was a very significant difference between them( P〈0. 01). The number of Pyramidal layer of cells and receptor positive neurons in the hippocampus and cerebral cortex M1 have increased,and there was a significant difference( P〈0. 01) in the CA1 and CA3 region and S1 Tr.Conclusion: yizhi jiannao granule can improve the abilities of learning and memory in aged rats,probably by enhancing the expression level of the basal forebrain cholinergic neurons Ch AT and increasing the amount of acetylcholine synthesis; Cholinergic muscarinic M1 receptor expression regulation may accept more projection from the basal forebrain,and triggered effects receptor,as to have a protective effect to cholinergic system in aged rats.
