经典型霍奇金淋巴瘤p16和p53蛋白表达与EB病毒感染相关性研究 被引量:12

Expression of p16 and p53 protein and association with epstein-barr virus infection in classic Hodgkin's lymphoma

摘要 目的 EB病毒(EB virus,EBV)感染、p16和p53蛋白的表达异常可能是经典型霍奇金淋巴瘤(classical Hodgkin’s lymphoma,CHL)发生和发展的危险因素,本研究探讨p16和p53蛋白在CHL中的表达及与EBV感染的相关性。方法选取1995-09-01-2014-06-30新疆自治区人民医院(91例)和新疆生产建设兵团第一师医院(17例)病理诊断为CHL的存档蜡块108例,应用R-S细胞中EBV编码小RNA(EBER-1)寡核苷酸探针以原位杂交(ISH)方法检测CHL细胞中EBER-1的表达;免疫组化方法检测CHL细胞中p16和p53蛋白的表达。并分析p16和p53蛋白与EBER-1表达的相关性。结果 CHL中EBER-1的阳性表达率为54.63%(59/108),其中淋巴细胞为主型(LR)阳性率为43.48%(10/23),混合细胞型(MC)为67.92%(36/53),结节硬化型(NS)为33.33%(7/21),淋巴细胞消减型(LD)为54.55%(6/11),差异有统计学意义,P<0.05。p16蛋白的阳性表达率为45.37%(49/108),其中LR阳性率为34.78%(8/23),MC为58.49%(31/53),NS为23.81%(5/21),LD为45.45%(5/11),差异有统计学意义,P<0.05。p53蛋白阳性表达率为64.81%(70/108),其中LR阳性率为60.87%(14/23),MC为73.58%(39/53),NS为42.86%(9/21),LD为72.73%(8/11),差异无统计学意义,P>0.05。EBV与p16蛋白的表达呈负相关,r=-0.256,P=0.008;EBV与p53蛋白的表达呈正相关,r=0.313,P=0.001。结论 CHL中存在EBV感染,且p16和p53高表达;EBV感染在p16和p53蛋白的表达中可能扮演了重要角色。 OBJECTIVE EB virus infection, the expression of p16 and p53 protein abnormalities may be the typical Hodgkin's lymphoma risk factors of the occurrence and development. The objective of this study is to study the relationship between the expression of p16, p53 protein and EB virus infection in classical Hodgkin lymphoma (CHL). METHODS One hundred and eight patients from 1995-09-01 to 2014-06-30 in First Division Hospital of Xinjiang Production and Construction Corps and Xinjiang Uygur Autonomous Region People's Hospital were included in the sample. EBER-1 and p16, p53 protein expression of CHL were studied with EBER-1 oligonucleotide probe and immunohisto chemistry, respectively. The correlation between p16, p53 protein and EBER-1 was analyzed. RESULTS The rate of EBER-1 posi tive expression in CHL was 54. 63% (59/108), the lymphoeytes mainly type (LR) was 43. 48% (10/23), mixed type cells (MC) was 67.92% (36/53), nodular sclerosis type (NS) was 33.33% (7/21), the lymphocytes destructive (LD) was 54.55%(6/11), there was statistically significant difference between the groups (P〈0.05). The rate of pl 6 protein expression inCHL was 45. 37%(49/108), LR positive rate was 34. 78%(8/23),MC was 58. 49%(31/53),NS was 23.81%(5/21),LD was45.45%(5/11), there was statistically significant difference between the groups (P〈0.05). The rate of p53 protein expression in CHL was 64. 81% (70/108), LR positive rate was 60. 87%(14/23), MC was 73.58%(39/53), NS was 42.86% (9/21), LD was 72. 73% (8/11), there was no statistically significant difference between the groups (P〉0.05). The rate of EBER-1 positive showed negative correlation with p16 protein(r= -0. 256, P=0. 008) ,but that showed positive correlation with p53 protein(r=0. 313, P=0. 001). CONCLUSION EB virus infection plays an important role in the expression of p16, p53 protein of the classical Hodgkin's lymphoma.
