摘要
目的 评价脂联素对小鼠肾脏缺血再灌注损伤的影响.方法 健康雄性C57BL/6小鼠32只,8周龄,体重20~25 g,采用随机数字表法,将其分为3组(n=8):假手术组(S组)、肾脏缺血再灌注组(I/R组)和脂联素+肾脏缺血再灌注组(A+I/R组).采用阻断双侧肾蒂30 min再灌注的方法制备肾脏缺血再灌注损伤模型.A+I/R组在缺血前15 min、再灌注24 h时分别腹腔注射重组小鼠脂联素球状结构域5 μg/g.于再灌注48 h时摘除眼球,取血样,检测血清BUN和Cr的浓度,随后取双侧肾脏,HE染色,光学显微镜下观察肾组织病理学结果,行肾小管损伤评分,并采用RT-PCR法检测IL-1β、IL-6和TNF-α mRNA的表达.结果 与S组比较,I/R组和A+I/R组血清BUN和Cr的浓度、肾小管损伤评分升高,肾组织IL-1β、IL-6和TNF-α的mRNA表达上调(P<0.05);与I/R组比较,A+I/R组血清BUN和Cr的浓度、肾小管损伤评分降低,肾组织IL-1β、IL-6和TNF-α的mRNA表达下调(P<0.05).A+I/R组肾组织病理学损伤明显轻于I/R组.结论 脂联素可减轻小鼠肾脏缺血再灌注损伤,其机制与抑制炎性反应有关.
Objective To evaluate the effect of adiponectin on renal ischemia-reperfusion (I/R) injury in mice.Methods Thirty-two male C57BL/6 mice, aged 8 weeks, weighing 20-25 g, were randomly divided into 3 groups (n =8 each) using a random number table: sham operation group (group S),I/R group and adiponectin + I/R group (group A+I/R).Bilateral kidneys were exposed, and their pedicles were occluded for 30 min with atraumatic miniclamp followed by reperfusion.Globular domain of adiponectin 5 μg/g was injected intraperitoneally at 15 min before ischemia and 24 h of reperfusion in group A+I/R.The mice underwent eye enucleation at 48 h of reperfusion, and blood samples were collected for determination of serum blood urea nitrogen (BUN) and creatinine (Cr) concentrations.Bilateral kidneys were obtained for microscopic examination and for determination of the expression of interleukin-1beta (IL-1β), IL-6 and tumor necrosis factor-alpha (TNF-α) mRNA expression (using real-time reverse transcriptase polymerase chain reaction).The damage to the renal tubules was scored.Results Compared with group S, the serum BUN and Cr concentrations, and renal tubular damage score were significantly increased, and the expression of IL-1β, IL-6 and TNF-α mRNA was up-regulated in I/R and A+I/R groups (P〈 0.05).Compared with group I/R, the serum BUN and Cr concentrations, and renal tubular damage score were significantly decreased, and the expression of IL-1β, IL-6 and TNF-α mRNA was down-regulated in group A+I/R (P 〈0.05).The pathological changes of renal tissues were significantly reduced in group A +I/R as compared with group I/R.Conclusion Adiponectin can mitigate renal I/R injury in mice, and the mechanism is related to inhibition of inflammatory responses.
