恐伤孕鼠对其仔鼠大脑皮质Reelin、VLDLR及P-DAB1表达的影响 被引量:6

Effects of horror injuring pregnant rats on the expression of cerebral cortex Reelin, VLDLR and P-DAB1 of filial rats

摘要 目的:研究恐伤孕鼠对其仔鼠大脑皮质Reelin、VLDLR及P-DAB1表达的影响。方法:将受孕后雌鼠用随机分为正常组、模型组、补肾组,每组各10只。运用恐吓为主的复合应激法刺激造模,正常组不予刺激,造模时间为自分组第1天直至产下仔鼠。补肾组灌胃中药左归丸浓缩剂,2m L/100g体质量/d,正常组及模型组则根据雌鼠体质量予以等量0.9%氯化钠溶液灌胃。仔鼠出生当天每窝随机取1-2只仔鼠断头取全脑,后将全脑固定,切取包含皮质的脑片。结果:模型组仔鼠大脑皮质区光镜下可见细胞较松散,组织结构较模糊,存在散在片状水肿,水肿区域神经细胞数目减少;补肾组与正常组比较,仔鼠大脑皮质在形态学上无明显改变。与正常组比较,补肾组和模型组仔鼠大脑皮质Reelin、VLDLR及P-DAB1的蛋白表达均下调(P<0.01);与模型组比较,补肾组仔鼠大脑皮质Reelin、VLDLR及P-DAB1的蛋白表达明显上调(P<0.01)。结论:恐伤孕鼠可以引起仔鼠大脑皮质发育异常,而左归丸通过调节Reelin信号通路可阻断或降低仔鼠大脑皮质发育障碍损伤的发生。 Objective: To study the impact of horror injuring pregnant rats on the expression of the cerebral cortex Reelin, VLDLR and P-DAB1 of filial rats. Methods: Pregnant rats were randomly divided into normal group, model group, kidney invigorating group with 10 rats in each group. Compound stress method of intimidation was mainly used to stimulate the model group, and the normal group was not received any stimulation. Modeling period was from the first day of grouping to the day giving birth to filial rats. The kidney invigorating group was received intragastric administration of concentration agent of Zuogui Pills with 2m L/100 g body weight/d. The normal group and model group were received intragastric administration of 0.9% sodium chloride solution with equal amount according to the weight of female rats. One or two filial rats were randomly selected to collect the whole brains on the day when the filial rats were born from each of group. Then the whole brains were fixed, and the brain slices contained with cortex were collected. Results: It was seen in cortical areas of filial rats in model group under light microscope that relatively incompact cells, vague tissue structure, scattered flake edema, and reduced amount of nerve cells in edema area. Compared with the normal group, cerebral cortex of filial rats in kidney invigorating group had no significant changes in morphology. Compared with normal group, all protein expressions of Reelin, VLDLR and P-DAB1 in filial rats' cerebral cortex in kidney invigorating group and model group were decreased(P<0.01); Compared with model group, protein expressions of Reelin, VLDLR and P-DAB1 in filial rats' cerebral cortex in kidney invigorating group were significantly increased(P<0.01). Conclusion: Horror injuring pregnant rats can cause the abnormal development of cerebral cortex of filial rats, and Zuogui Pills can block or reduce the occurrence of injury development of cerebral cortex of filial rats by adjusting Reelin signal pathways.
