摘要
目的:研究骨痹颗粒对骨关节炎(OA)相关炎性反应的作用,探讨其作用机制。方法:建立小鼠耳肿胀模型和大鼠棉球肉芽肿模型,观察骨痹颗粒对小鼠耳肿胀度及大鼠肉芽质量、血清前列腺素E2(PGE2)含量的影响;建立大鼠骨关节炎模型,检测各组大鼠血清中白细胞介素6(IL-6)、PGE2含量,诱导型-氧化氮合酶(i NOS)表达及NO水平。结果:骨痹颗粒可明显降低小鼠耳廓肿胀度(P<0.01),降低大鼠棉球肉芽肿系数及血清PGE2的水平(P<0.01,P<0.05);减少OA大鼠血清IL-6、PGE2水平,下调i NOS表达,降低NO生成(P<0.01,P<0.05)。结论:骨痹颗粒能够抑制非特异性急、慢性炎性反应及骨关节炎特异性炎性反应,减少相关炎性介质及炎性因子的释放而抑制相关炎性反应的发生与发展。
Objective: To study the effects of Gubi Granule on osteoarthritis-related inl ammation reaction and explore its mechanism. Methods: The mice model of ear swelling and the rat model of cotton ball granuloma were established to observe the effects of Gubi Granule on ear swelling, the granuloma weight and the content of PGE2 in serum. The rat model of osteoarthritis was established to determine the contents of IL-6, PGE2, fiNOS and NO in serum. Results: Gubi Granules significantly inhibited ear swelling of mice(P〈0.01). It also decreased granuloma coei cient in rats and reduced the level of PGE2 in the rat serum(P〈0.01, P〈0.05). Meanwhile, Gubi Granules significantly reduced the levels of IL-6, PGE2, fiNOS and NO in serum of osteoarthritis rats(P〈0.01, P〈0.05). Conclusion: Gubi Granules could suppress not only the acute or chronic nonspecific inl ammation reaction but also specific inl ammation reaction of osteoarthritis, which might be closely related to inhibit the release of inl ammatory mediators and factors.
