摘要
目的:探讨补肾壮骨滴丸中主要活性成分淫羊藿苷在骨质疏松模型与正常大鼠体内组织分布的差异。方法:SPF大鼠分为骨质疏松模型组和正常组,各分为5组,每组6只。各组按淫羊藿苷118 mg·kg-1灌胃给予补肾壮骨滴丸。于40 min,1.5,4,24 h取各组织,采用高效液相色谱法测定各组大鼠各组织中淫羊藿苷及其代谢产物淫羊藿次苷Ⅰ、淫羊藿次苷Ⅱ、淫羊藿素的质量浓度。结果:补肾壮骨滴丸中淫羊藿苷经口给药后,在模型组大鼠体内吸收分布较正常组稍缓,但蓄积浓度较高,体内存留时间延长;代谢产物在模型大鼠组织中分布浓度也高于正常大鼠组,体内停留时间较长,其中淫羊藿素在肌肉组织中的浓度远远高于正常大鼠组,具显著性差异。结论:本实验通过比较正常及病理状态下经口服给药后,从体内组织分布方面证明了药物在不同环境下体内动力学特征存在差异。这可能与本方补肾填精同时,激发肾精的组织修复功能存在某种内在的联系,有待于进一步的研究。
Objective: To compare the tissue distribution of icariin in Bushen Zhuanggu Dripping Pills between model and normal rats. Method: The osteoporosis model group and the normal group were both divided into 5 groups( n = 6).Tissues were collected after oral administration of 118 mg·kg-1icariin at 40 min,1. 5,4,24 h. The concentration of each component was determined by HPLC after sample preparation. Result: Icariin was distributed more slowly in model group than that in normal group,but had higher concentration and longer lifetime than that in normal group after oral administration of Bushen Zhuanggu Dropping Pills. Conclusion: It is proved that there is a difference in the kinetics of the drug in different environments between the model group and the normal group. This may be related to the Bushen Tianjing and at the same time stimulating tissue repair with intrinsic link,which needs further study.
