Ccbe1病毒对喉癌Hep-2细胞VEGF-C表达的影响

摘要 目的探讨胶质和钙结合EGF区域1(Collagen and calcium binding EGF domains 1,Ccbe1)病毒对人喉癌细胞系Hep-2细胞增殖及血管内皮生长因子-C(vessel endothelium growth factor C,VEGF-C)表达的影响。方法用1×102,1×104,1×106 pfu/m L的Ccbe1腺病毒作用喉癌Hep-2细胞,细胞分为对照组(PBS),Ccbe1病毒高、中、低剂量组。观察各组细胞增殖情况,采用PCR检测各组细胞VEGF-C mRNA的表达水平。结果与对照组比较,Ccbe1病毒各剂量组细胞增殖显著增高,VEGF-C mRNA表达水平显著上升,且均有显著剂量依赖性,差异有统计学意义(P<0.05)。结论 Ccbe1病毒可能通过上调淋巴管生成因子VEGF-C表达促进喉癌细胞增殖和淋巴管生成。 Objective To investigate the impact on Vascular endothelial growth factor C(VEGF-C) expression of Collagen and calcium-binding EGF domain-containing protein 1(Ccbe1) virus to laryngeal cancer Hep-2 Cells. Methods Cells were divided into control group(PBS), Ccbe1 virus in high, medium and low dose group, by the way of Ccbe1 adenovirus(in respective units 1 ×10^2, 1×10^4, 1×10^6pfu/mL) acting on laryngeal cancer Hep-2 Cells. The cell proliferation was observed and the expression level of VEGF-C m RNA was detected by PCR for each group. Results Compared with the control group, the cell proliferation of Ccbe1 virus was significantly higher in each group, and the VEGF-C m RNA expression level was significantly increased than that in control group(P〈0.05), and there were significant dose-dependence. Conclusion Ccbe1 virus may upgrade Lymph vessel generation factor VEGF-C expression to promote hep-2 cells proliferation and lymphatic formation in laryngeal cancer.
