NF-κB对肺动脉高压大鼠肺动脉组织前列环素表达的影响 被引量:2

摘要 目的观察核转录因子κB(NF-κB)对肺动脉高压(PH)大鼠肺动脉组织前列环素(PGI2)表达的影响。方法将50只大鼠随机分为4组,Tn、Ti组各15只,均通过颈总动脉—颈外静脉吻合建立左向右分流型PH模型,Ti组术前1 h腹腔注射NF-κB阻断剂吡咯烷二硫基甲酸盐(PDCT)120 mg/(kg·d),术后持续2周;Co组10只除不给予颈总动脉—颈外静脉吻合外其余造模过程与Tn、Ti组相同,Cn组10只不处理。各组连续饲养12周后,采用心导管术测量右心室收缩压代表肺动脉收缩压(PASP),通过凝胶迁移率实验检测肺动脉内皮细胞NF-κB活性,以免疫组化法检测肺动脉组织中的PGI2。结果 Tn组肺动脉内皮细胞NF-κB活性高于Cn组(P<0.01),Ti组NF-κB活性低于Cn组(P<0.01),而Co组与Cn组比较无明显差异。Tn组大肺动脉组织PGI2表达量较Cn组明显减少(P<0.01),Ti组与Cn组、Co组与Cn组比较均无明显差异。结论 PH大鼠肺动脉内皮细胞NF-κB活性增加,可抑制血管活性物质PGI2的表达。
