摘要
目的探讨细胞外信号调节激酶(Extracellular Signal-regulated Kinase,ERK1/2)信号通路在食管癌中的功能及其对上皮间质转化(Epithelial-Mesenchymal Transition,EMT)的调控作用。方法实验组使用ERK1/2磷酸化抑制剂U0126,抑制ERK1/2的磷酸化程度后,阴性对照组为正常培养的Eca109细胞,采用噻唑蓝实验、细胞划痕以及Transwell实验分别检测ERK1/2对食管癌细胞Eca109增殖、迁移和侵袭的影响,通过Western blot实验检测ERK1/2对EMT相关蛋白Vimentin表达的影响。结果 U0126作用后,食管癌细胞Eca109增殖受到抑制,迁移速度减慢,侵袭细胞数显著减少,同时Vimentin蛋白表达量显著降低。结论磷酸化的ERK1/2能够促进食管癌细胞的增殖、迁移和侵袭;磷酸化的ERK1/2能够调控Vimentin的表达,食管癌中ERK1/2可能参与了EMT过程,从而促进了食管癌细胞的侵袭和转移。
Objective To investigate the role of extracellular signal-regulated kinase( ERK1 /2) and its regulation of epithelial-mesenchymal transition( EMT) in esophageal squamous cell carcinoma( ESCC). Methods The experiment group was performed using U0126,and the negative control group was normal cultured Eca109 cells. The effective of ERK1 /2 on proliferation,migration and invasion of ESCC cell Eca109 were tested by MTT,wound healing and transwell after dephosphorylated ERK1 /2. Expression of vimentin was detected by western blot. Results The proliferation of Eca109 cell was inhibited,the rate of migration was slowed down,and the number of invasive cells was significantly reduced after deal with U0126. Meanwhile,the expression of vimentin was significantly reduced. Conclusion Phosphorylated ERK1 /2 contributed to the proliferation,migration and invasion of ESCC cells. Phosphorylated ERK1 /2 regulated the expression of vimentin,it may be involved in the EMT process,thus contributing to the invasion and metastasis of ESCC.
