脱氢紫堇碱对高糖培养心肌成纤维细胞Wnt信号通路的影响 被引量:2

摘要 目的采用体外高糖培养心肌成纤维细胞(CFs)的方法,观察脱氢紫堇碱(DHC)对高浓度葡萄糖培养的心肌成纤维细胞Wnt信号通路的影响。方法采用体外高糖诱导心肌成纤维细胞增殖模型,western blot法检测β-catenin,GSK-3β以及P-GSK-3β的蛋白表达水平;ELISA法观察细胞分泌TGF-β1蛋白的能力。结果脱氢紫堇碱可以抑制β-catenin的表达(P<0.01),上调P-GSK-3β的表达(P<0.05),减少TGF-β1的合成(P<0.01)。结论脱氢紫堇碱能抑制Wnt信号通路的异常表达,具有潜在的抗心肌纤维化作用。 Objective Take the method of cultured cardiac fibroblasts (CFs) in vitro, to observe the effect of Dehydrocorydaline on high glucose cultured cardiac fibroblasts of Wnt signal pathway. Methods By the cardiac fibroblasts model in vitro induced by high glucose, detection of the expression level of β-catenin, GSK-3β, P-GSK-3β by western blot ; to observe the ability of the cell TGF-β1 protein secretion by ELISA method. Results Dehydrocorydaline can inhibit the expression of β-catenin (P 〈 0.01 ), upregulate the expression of P-GSK-3β(P 〈 0. 05 ), reduction of the synthesis of TGF-β1 (P 〈 0.01 ). Conclusion Dehydrocorydaline can inhibit of abnormal expression of Wnt signaling pathway , have potential effects of anti myocardial fibrosis
