磁珠联合MALDI-TOF-MS筛选尿液中早期肾透明细胞癌多肽标志物

摘要 目的寻找尿液中潜在的肾癌多肽标志物。方法收集肾癌患者根治性肾切除术前、术后组、非肾癌对照组晨起第二次中段尿液标本。铜螯合纳米磁珠提取尿多肽,MALDI-TOF-MS分析各组尿液中肾癌相关多肽标志物,并通过Tagldent程序对有意义多肽进行初步鉴定。结果 m/z=1 025、1 116的多肽在术前组高表达,在术后组、对照组低表达。m/z=9 730的多肽在术后组、对照组高表达,在术前组低表达。初步鉴定发现,m/z=1 025信号峰与Urotensin-2B、Little LEN匹配,m/z=1 116信号峰与GRP匹配,m/z=9 730信号峰与EA-92、Late cornified envelope protein 3C、Heparin-binding EGF-like growth factor匹配。结论肾癌患者尿液中高表达m/z为1 025、1 116的多肽,低表达m/z为9 730的多肽,这些多肽可能是尿液中潜在的肾癌标志分子。 Objective To investigate potential urinary pcptide markers for early stage clear cell renal cell carcinoma (ccRCC). Methods Middle stream samples of 2"d morning urine from RCC patients were collected pre-operatively and 7 days post-operatively. Urine samples from no-RCC patients were used as control. Urine peptides were separated by Cuprum chelated magnetic bead, and then analyzed by MALDI-TOF-MS. Tagldent program was employed to identify RCC related peptides. Results Peptides with m/z=1025 and m/z=1 116 were up-expressed in pre-operative RCC patients, and down-expressed post-operatively and in controls. Peptides with m/z= 9 730 was up-expressed in post-operative RCC patients and in controls. Urotensin-2B and Little LEN matched peptide with m/z = 1 025, GRP matched peptide with m/z = 1 116, EA-92, Late corni- fied envelope protein 3C and HB-EGF matched peptide with m/z=9 730. Conclusion Peptides with m/z=1 025 and m/z = 1 116 were up-expressed in urine of ccRCC patients, while peptide with m/z = 9 730 was down-expressed. These peptides may be important potential urinary tumor markers for ccRCC.
