摘要
目的:探讨南蛇藤提取物(Celastrus orbiculatus extract,COE)对人胃癌SGC-7901细胞在裸鼠体内转移的影响及其可能机制。方法:建立裸鼠腹腔转移瘤模型,随机分为对照组,COE低剂量组(10 mg·kg-1)、中剂量组(20 mg·kg-1)、高剂量组(40mg·kg-1),阳性对照组(希罗达)。观察COE对人胃癌SGC-7901细胞在裸鼠体内转移的影响。应用免疫组化法检测腹腔转移瘤组织中E-钙黏蛋白(E-cadherin)、N-钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)的表达。结果:COE各剂量组能够明显抑制腹腔转移瘤的生长、腹膜种植转移和肝肺转移的发生(P<0.05),且呈剂量依赖效应。COE处理后腹腔转移瘤组织中Ecadherin蛋白表达有升高的趋势,而N-cadherin和Vimentin蛋白表达有降低的趋势。其中与对照组相比,COE高剂量组中Ecadherin蛋白表达显著升高(P<0.05),而N-cadherin和Vimentin蛋白表达显著降低(P<0.001)。结论:COE对人胃癌细胞SGC-7901在裸鼠体内的转移具有明显的抑制作用,其机制可能与增高E-cadherin的表达,降低N-cadherin和Vimentin的表达,抑制胃癌EMT的发生有关。
Objective: To investigate the effects and mechanisms of Celastrus orbiculatus extract( COE) on metastasis of human gastric carcinoma SGC-7901 cells in vivo. Method: SGC-7901 cells were inoculated intraperitoneally in BALB / C nude mice. Two weeks after inoculation,these mice were randomly divided into negative control group( 1% DMSO),positive control group( xeloda),and low,medium and high dose COE groups( 10,20 and 40 mg·kg- 1,respectively). The anti-metastasis effects of COE on nude model mice of gastric cancer were observed and the mechanisms were explored. The expression of EMT markers were evaluated with immunohistochemistry. Results: In COE dose groups the growth of metastases of the abdominal cavity,the occurrence of peritoneal planting metastasis and liver-lung metastasis could be significantly inhitited( P〈0. 05),with dose dependent model. Immunohistochemical results suggested that E cadherin protein expression in retroperitoneal metastatic tumor tissue had a tendency to rise after being processed by COE,while N-cadherin and Vimentin protein expression had tendencys to reduce. Compared with the control group,the E-cadherin protein expression of COE high doese group significantly increased( P〈0. 05),while the N-cadherin and Vimentin protein expression significantly reduced( P〈0. 001). Conclusion: COE is an effective inhibitor of metastatic potential of SGC-7901 cells through suppression of EMT.The mechanism may be related to the increasing expression of E-cadherin,the decreasing expression of N-cadherin and Vimentin,with function to inhibite the pathogenesis of EMT of gastric cancer.
