摘要
目的探讨氯沙坦对静态牵张刺激导致的肥大心室肌细胞中L-型钙离子通道Cav1.2亚基及其蛋白表达的影响。方法 1d龄SD乳鼠,采用胰酶消化法分离获得心室肌细胞分为对照组、静态牵张组、氯沙坦组、静态牵张+氯沙坦组。定量分析细胞蛋白/DNA比值、BNP及细胞面积以鉴定牵张有效性。实时荧光定量PCR检测Cav1.2的mRNA水平;蛋白质印迹法检测Cav1.2蛋白表达水平。结果静态牵张刺激导致心室肌细胞蛋白/DNA比值,BNP基因表达,心室肌细胞面积增大。静态牵张刺激导致心室肌细胞Cav1.2mRNA及蛋白表达水平下调,氯沙坦干预该效应。结论氯沙坦可以抑制静态牵张刺激导致的心室肌细胞Cav1.2基因及蛋白水平的低表达。
Objective To explore the effects of losartan on Ca-Lion channel stimulated by stretch in ventricular myocyte from neonatal rats.Method Neonatal rat ventricular myocyte was isolated and cultured into control group,stretched group,losartan group and stretched+losartan group.Alterations of protein-to-DNA ratio,brain natriuretic peptide levels and cell size level in the cell culture supernatant were used to determine the effectiveness of in vitro stretched model.The mRNA and protein expression levels of Cav1.2were assayed by real-time PCR and western blot(WB)test.Result Stretch significantly increased the protein-to-DNA ratio,BNP and cell size.The transcription of Cav1.2mRNA and expression of its proteins in stretched group were significantly reduced,which was prevented by losartan.Conclusion Losartan may prevent the lower expression of Cav1.2in mRNA and protein induced by stretched.
