摘要 目的研究CAMKK2在前列腺癌及癌旁组织中的表达,分析其表达与临床病理数据的关系。方法通过实时荧光定量PCR技术检测CAMKK2的mRNA水平在前列腺癌及癌旁组织中的表达情况,通过免疫印迹及免疫组化技术检测CAMKK2的蛋白水平在两种组织中的表达,分析CAMKK2的表达水平与前列腺癌相关临床病理特征及预后的关系。结果前列腺癌组织中的CAMKK2在mRNA及蛋白水平上均较癌旁组织中上调表达,CAMKK2的表达与血清PSA水平、是否转移及是否生化复发有关,CAMKK2的表达高低与无生化复发生存率相关,CAMKK2是生化复发的独立危险预测因素。结论 CAMKK2在前列腺癌组织中的表达增加,可能提示患者的预后情况,并可作为前列腺癌新的生物标记物。 Objective To investigate the expression and significances of CAMKK2 in prostate cancer and adjacent non-cancerous tissues,and analyze the associations of the expression of CAMKK2 and the clinical pathological features.Method Q-PCR was used to detect the CAMKK2 mRNA level in prostate cancer and adjacent non-cancerous tissues,western blot(WB)and immunohistochemistry(IHC)were used to detect the CAMKK2 protein level,the associations of CAMKK2 expression with prostate cancer clinical pathological features and prognosis were analyzed.Result The mRNA and protein level of CAMKK2 were upregulated in prostate cancer tissues than in adjacent non-cancerous tissues,the expression of CAMKK2 had relation with serum PSA level,metastasis and PSA failure,the expression of CAMKK2 had relation with biochemical non-recurrence survival,and it might be an independent predictor of biochemical recurrence in prostate cancer.Conclusion The upregulation of CAMKK2 in prostate cancer may predict the prognosis of prostate cancer patient,and it may be a new biomarker of prostate cancer.
