微生物教学实验中菌株革兰氏染色反应快速判定 被引量:6

摘要 依据荧光标记的凝集素能够与革兰氏阳性菌细胞壁肽聚糖中的N-乙酰葡糖胺结合,却无法透过革兰氏阴性菌的细胞壁外膜与革兰氏阴性菌肽聚糖层中的N-乙酰葡糖胺结合的特点,提出快速判定菌株革兰氏染色反应的荧光法。针对供试的27株菌,该法革兰氏染色反应判定的准确率高达98.76%,且不受菌龄影响。设置阳性对照抑制实验,有利于提高判定准确率。与常规革兰氏染色法相比,该法具有所需药品少、简单、快速、准确率高的特点,尤其在不可培养样品的革兰氏染色反应判定方面具有很大的应用前景。在微生物教学中可以考虑将该法与传统革兰氏染色法联合使用,以提高教学效果。 According to the characteristics that fluorescent-labeled wheat germ agglutinin capable of binding to N-acetylglucosamine in the out peptidoglycan layer of gram-positive bacteria,which could not penetrate the membrane of gram-positive bacteria to bind to N-acetylglucosamine,a fluorescent stain method using flurescent lectin is developed for the fast determination of Gram-stain reaction.For the 27 strains tested,the accuracy of developed method is as high as 98.76%and is not affected by the cell age.Setting positive control inhibition experiments could help to improve the accuracy of the developed method.Compared with the conventional Gram staining,this method has the characteristics of requiring fewer types of drugs,simple,fast and high accuracy,it has great application prospects especially in non-cultured samples.Therefore,in the teaching of microorganisms,it is possible to combine this method with traditional Gram staining to improve the teaching effectiveness.
