碘酸钠诱导兔视网膜色素上皮变性模型的研究 被引量:2

摘要 目的通过静脉注射不同剂量的碘酸钠后观察兔视网膜色素上皮形态学及视网膜功能变化,以探索注射碘酸钠的合适剂量。方法选取成年健康无眼病青紫兰兔32只,随机分为对照组5只,实验1组(静脉注射碘酸钠15 mg·kg-1)、实验2组(静脉注射碘酸钠25 mg·kg-1)、实验3组(静脉注射碘酸钠35 mg·kg-1)各9只。对照组和实验组分别在耳缘静脉注射碘酸钠后1 d、3 d、7 d、15 d、28 d视网膜电图检查后处取视网膜后极部进行光镜及透射电镜观察。结果实验2、3组诱导了兔视网膜色素上皮变性。实验2组:给药后3 d可见大量的视网膜色素上皮细胞自Bruch膜上脱落,视细胞外节破坏,游离入感光细胞层;给药后7 d可见更严重的感光细胞破坏溶解,但外界膜完整;随时间推移;给药后28 d外核层细胞数明显减少,视网膜成为无视网膜色素上皮或多层视网膜色素上皮相间结构。实验3组:给药后3 d即可见大量的视细胞内外节溶解坏死,外界膜、外核层受损。结论静脉注射适当剂量的碘酸钠生理盐水能够选择性破坏视网膜色素上皮,成功造成视网膜色素上皮变性模型,25 mg·kg-1碘酸钠为合适剂量。 Objective To observe the changes of retinal pigment epithelial mor- phology and retinal function after intravenous injection of sodium iodate with different doses, and study the proper dose of sodium iodate to induce the pigment epithelial de- generation. Methods Thirty-two adult healthy rabbits were randomly divided into control group( 5 cases), experimental group 1 (intravenous injection of 15 mg·kg^-1 so- dium iodate,9 cases), group 2 (intravenous injection of 25 mg·kg^-1 sodium iodate, 9 cases), group 3 ( intravenous injection of 35mg·kg^-1 sodium iodate,9 cases). ERG was used before all the rabbits were sacrificed at 1 day,3 days,7 days,15 days and 28 days after the treatment. Retinas were examined histologically and electrophysiologically. Results There was no pathology change in group 1 after iodate sodium treated. Reti- nal pigment epithelial degeneration was evoked in group 2 and group 3. In group 2, the primary site of damage appeared in RPE cells, followed by photoreceptor outer segment disruption at 3 days, the RPE detached from the Bruch membrane and migrated to the inner retina;At 7 days, more photoreceptor were disrupted and the inner limiting was complete;At 28 days, the retina showed a mosaic pattern of relatively normal retina and areas lacking RPE cells. In group 3, the destruction of retina was more severe, the outer nuclear layer was already damaged at 3 days. Conclusion Intravenous injection of sodium iodate can selectively damage RPE and followed by photoreceptors degenera- tion. The proper dose of sodium iodate to induce the retinal pigment epithelial degenera- tion is 25 mg·kg^-1.
