摘要
为了对用于检测牛奶中阿莫西林残留的酶联免疫吸附试验(ELISA)检测试剂盒进行质量验证,试验采用不同梯度浓度的方法进行研究。结果表明:阿莫西林标准品浓度在0.5-13.5μg/L范围内具有较好线性关系,50%竞争抑制浓度为2.0μg/L,试剂盒对牛奶中阿莫西林的最低检测限为4.6μg/L。牛奶中分别添加5.0,10.0μg/L的阿莫西林标准品,回收率在72.2%-108.7%之间,批内变异系数小于8.9%,批间变异系数小于11.9%;试剂盒可在37℃条件下保存7 d,可同时检测牛奶中阿莫西林、青霉素、氨苄西林、萘夫西林等药物的残留总量。与仪器方法进行实际样品检测相比,ELISA方法检测结果具有良好的可信度。
To perform quality verification of enzyme - linked immunosorbent assay (ELISA) detection kit for amoxicillin residue in milk, a method of different gradient concentrations was used in the test. The results showed that the standard concentration of amoxicillin exhibited a good linearity in the range of 0.5 to 13.5 μg/L with the 50% competitive inhibition concentration of 2.0 μg/L. The lowest detection limit of the kit for amoxieiUin in milk was 4.6 μg/L. When adding standard amoxicinin of 5.0 and 10.0 μg/L, respectively, the recovery rates were between 72.2% to 108.7%, and the coefficient of variations (CV) for intra - assay and inter - assay were less than 8.9% and 11.9%, respectively. The kit could be saved in 37℃ for 7 days, and suitable for the determination of total residues of amoxicillin, penicillin, ampieillin and nafeinin in milk. ELISA method had a good reliability for detecting practical samples compared with instrumental method.
