摘要
目的分析胰岛素样生长因子Ⅱ(insulin-like growth factorⅡ,IGF-Ⅱ)及胰岛素样生长因子ⅡmRNA结合蛋白3(insulin-like growth factorⅡmRNA binding protein 3,IMP3)在宫颈组织中的表达及意义。方法收集2013年1月至2015年3月德州市人民医院妇科就诊患者取活检或者手术切除后的宫颈组织标本85例,其中宫颈癌42例,正常宫颈组织15例,宫颈炎10例,宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)Ⅰ期7例、Ⅱ-Ⅲ期11例,采用免疫组化的方法检测组织中IGF-Ⅱ及IMP3蛋白表达情况,对不同宫颈组织标本的IGF-Ⅱ及IMP3蛋白阳性表达率进行比较,并分析IGF-Ⅱ及IMP3蛋白的表达与宫颈癌临床病理特征之间的关系。结果不同宫颈组织标本的IGF-Ⅱ蛋白阳性表达率不全相等(P<0.05):CINⅡ-Ⅲ期组及宫颈癌组的IGF-Ⅱ蛋白阳性表达率高于正常组,宫颈癌组的IGF-Ⅱ蛋白阳性表达率高于宫颈炎组、CINⅠ期组及CINⅡ-Ⅲ期组(P<0.05)。不同宫颈组织标本的IMP3蛋白阳性表达率不全相等(P<0.05):宫颈癌组的IMP3蛋白阳性表达率高于正常组、宫颈炎组、CINⅠ期组及CINⅡ-Ⅲ期组(P<0.05)。不同年龄、人乳头瘤病毒(human papillomavirus,HPV)感染与否、肿瘤分化程度及不同病理类型之间患者的IGF-Ⅱ及IMP3蛋白的阳性表达率比较差异无统计学意义(P>0.05)。结论 IGF-Ⅱ及IMP3蛋白表达可为宫颈癌诊断提供一定的依据,可将其作为宫颈癌筛查的生物学标志物之一。
Objective To analyze the expression of Insulin - like growth factor Ⅱ mRNA binding protein 3 （ IMP3 ） and Insulin -like growth factor 1I （IGF -Ⅱ） in cervical tissue and its clinical significance. Methods The cervical tissues of 85 patients in Department of Gynecology of Peopled Hospital of Dezhou City from Jan 2013 to Mar 2015 were collected, included 15 cases of normal cervical tissues, 10 cases of cervieitis tissues, 7 cases of CIN I specimens, 11 cases of CIN Ⅱ - Ⅲ specimens, 42 cases of cervical cancer specimens. The positive expression rate of IGF - Ⅱ and IMP3 protein in different tissue samples detected by immunohistoehemistry were compared, and the relationship between IGF - Ⅱ and IMP3 protein expression and the clinical and patholigical features of cervical cancer were analyzed. Results The positive expression rate of the IGF - Ⅱ protein in different cervical tissue specimens were not equal （ P 〈 0. 05 ） , the positive expression rates of IGF - Ⅱ protein in CIN H - Ⅲ group and cervical carcinoma group were higher than that in normal group, the positive expression rates of IGF - Ⅱ protein in cervical carcinoma group was higher than that in cervicitis group, CIN I group, and CIN Ⅱ - Ⅲ group （P 〈 0. 05）. The positive expression rate of IMP3 protein in different cervical tissue specimens was not all equal （ P 〈 0. 05 ）, the positive expression rate of IMP3 protein in cervical cancer group was higher than that in normal group, cervicitis group, CIN I group and CIN Ⅱ -Ⅲ group （P 〈 0.05 ）. The difference in positive expression rates of IGF - Ⅱ and IMP3 protein between different ages, HPV infections, degrees of tumor differentiation and different pathological types had no statistical significance（P 〉 0. 05 ）. Conclusion IMP3 and IGF - Ⅱ expressions may provide a certain experimental evidence for pathological diagnosis of cervical cancer, which can be used as potential biomakers of cervical cancer screening.
