摘要
目的探讨3D平台在中国造血干细胞捐献者资料库(简称中华骨髓库)入库分型中的应用。方法基于3D仪器平台,使用试剂A对2014年中华骨髓库四川分库1 757份标本进行分型检测,使用Fusion3.4软件进行结果分析,依据中华骨髓库的入库标准指定结果,统计高分辨数据比例、中分辨模棱两可数据类型,并与四川分库2013年986份试剂B的分型结果和PCR-SBT方法的分型结果进行比较。结果 1 757例标本经试剂A分型后有3例为新等位基因,其余1 754例标本显示A、B、DR位点的高分数据百分比分别是98.5%(n=1 728)、96.5%(n=1 695)、99.2%(n=1 741),总的高分辨数据百分比95.1%(n=1 668),模棱两可结果 A、B、DR位点分别有5种、18种、8种;986例经试剂B分型显示A、B、DR位点的高分数据百分比分别是99.1%(n=977)、74.9%(n=739)、99.0%(n=976),总的高分辨数据百分比73.5%(n=725),模棱两可结果 A、B、DR位点分别有2种、83种、3种;1 177例PCRSBT试剂结果显示A、B、DR位点的高分数据百分比分别是92.9%(n=1 094)、95.9%(n=1 129)、96.6%(n=1 137),总的高分辨数据百分比86.8%(n=1 022),模棱两可结果 A、B、DR位点分别有10种、16种、6种。结论 3D平台支持的SSOP技术操作和数据分析简便可行,其高分辨分型结果能够满足目前中华骨髓库供者HLA入库分型的要求。
Objective To evaluate the feasibility of Luminex-3D based kits application in China Marrow Donor Program (CMDP) recruitment typing. Methods A total of 1757 donors recruited by CMDP Sichuan braneh in 2014 were typed by Luminex-3D based kit A, and the results were assigned by Fusion3.4 aeeording to CMDP standards. The percentage of high resolution and ambiguity types via kit A were counted, and also were compared to the results of 986 HLA assignments via kit B and 1174 assignments via PCR-SBT kit in 2013. Results By using kit A, 3 novel alleles were identified out of 1757 sam- pies. The percent of high resolution assignments in HLA-A ,-B, -DRB1 loci ware 98.'5% (n = 1 728), 96.5% (n = 1 693), 99.2% ( n = 1 741 ), respectively , and the total percent of high resolution assignments were 95.1% ( n = 1 668). And 5,18 and 8 kinds of ambiguous results were found in HLA-A, -B and -DRB1 typings, respectively. In 986 assignments via kits B, the pereem of high resolution assignments in HLA-A ,-B, -DRB1 loci ware 99. 1% (n = 977 ) ,74. 9% (n = 739) ,99.0% (n = 976), respectively , and the total percent of high resolution assignments were 73.5% ( n = 725 ). And 2, 83, 3 kinds of ambiguous results were found in HLA-A, -B and -DRB1 typings, respectively. Inl 177 assignments via PCR-SBT, the per- cent of high resolution assignments in HLA-A ,-B, -DRB1 loci ware 92.9% (n = 1094) ,95.9% (n = 1 129 ) and 96.6% (n = 1 137), respectively , and the total percent of high resolution assignments were 86. 8% ( n = 1 022). And 10,16,6 kinds of ambiguous results were found in HLA-A, -B and -DRB1 typings, respectively. Conclusion Luminex-based 3D PCR- SSOP kit are reliable and handy, and the typing resolutions were suitable for recruitment typing for CMDP.
