摘要
目的 了解2013-2014年监测年度我国B型流感病毒的流行特点,明确当前流行株与国际疫苗株和国内代表株的匹配情况及流感病毒的抗原性和基因特性变异情况.方法 选择2013年4月至2014年3月流感监测年度中国大陆分离的B型流感病毒,利用红细胞凝集抑制试验(HI)进行抗原分析.利用Sanger测序法进行病毒基因测序,采用MEGA 5.0软件进行我国B型流感病毒的种系进化变异分析.结果 2013年至2014年监测年度流行的B型流感病毒以B-Yamagata系流感病毒为主.B-Yamagata系流感病毒与本年度疫苗株B/Massachusetts/2/2012和我国代表株B/Chongqing-Yuzhong/1361/2013的抗原性类似.B-Victoria系流感病毒的抗原性与WHO推荐的疫苗株B/Brisbane/60/2008和我国代表株B/Beijing-Haidian/1386/2013的抗原性类似.基因特性分析显示102株B型流感病毒中,有60株(58.8%)病毒发生了B-Victoria系HA和B-Yamagata系NA间的重配.结论 本监测年度我国使用的疫苗株与我国流行的流感病毒匹配,我们所筛选的国内代表株能够代表我国各地流行株的病原学特征.
Objective To understand the epidemiological characteristics of influenza B virus,to analysis the matching of current circulating strains with international vaccine strains and national representative strains,and to analysis the antigenic and genetic variation of influenza B virus in year 2013-2014.Methods The Antigenic characteristics of influenza B virus was analyzed with hemagglutination inhibition (HI) assay.The nucleotide sequences of the influenza B viruses were determined by Sanger dideoxy sequencing.The phylogenetic trees were constructed by MEGA5.0 software,the genetic characteristics of the circulating influenza B virus were determined and compared with current vaccine strains.Results In 2013-2014 surveillance year,the activity of B-Yamagata lineage virus were higher than B-Victoria lineage virus.Most of the B-Yamagata lineage viruses were antigenically closely related to the WHO recommended vaccine strain B/Massachusetts/2/2012 and representative epidemic strain B/Chongqing-Yuzhong/1361/2013in China's Mainland.Vast majority of B-Victoria lineage viruses were antigenically closely related to vaccine virus B/Brisbane/60/2008 in the previous year and representative epidemic strain B/Beijing-Haidian/1386/2013;Among the 102 influenza B viruses,sixty of the B-Yamagata lineage viruses were confirmed to be an intra-lineage reassortants,with NA gene from B Victoria lineage.Conclusions The circulating influenza B virus in 2013-2014 was matched with the vaccine strain,and the representative viruses in China's Mainland were proved to be representing the characteristics of the epidemic strain.
