摘要
为探明脂蛋白脂酶(LPL)基因多态对鸭脂肪酸组成的影响,采用PCR-SSCP法和DNA直接测序相结合对鸭LPL基因内含子5和外显子8多态性进行检测,并分析其与鸭脂肪酸组成的关联性。结果表明:在内含子5中有4个SNP位点,分别位于243位(T→A)、244位(A→T)、265位(G→A)和269位(G→A);外显子8中有1个SNP位点,位于1 251位(T→C),为沉默突变。2个多态座位产生3种聚合基因型:AA/TT、AB/TC和BB/CC;经关联分析,等位基因A/T的脂肪酸月桂酸(C12:0)、豆蔻酸(C14:0)、棕榈酸(C16:0)、棕榈烯酸(C16:1)、十七烷酸(C17:0)、亚油酸(C18:2)、花生一烯酸(C20:1)、花生三烯酸(C20:3)、多不饱和脂肪酸(PUFA)和必需脂肪酸(EFA)含量与等位基因B/C间存在显著加性效应(P<0.05),不饱和脂肪酸(UFA)间存在显著显性效应(P<0.05)。等位基因A/T的脂肪酸组成更有利于人体健康。
The polymorphism of intron 5 and exon 8 of LPL gene was detected by PCR-SSCP and DNA direct sequencing and the relationships between their polymorphism and fatty acid composition were analyzed to discuss the effect of LPL gene on fatty acid composition of duck. Results: There are 4 SNP loci in intron 5 and they are located on 243 locus (T→A), 244 locus(A→T), 265 locus(G→A)and 269 locus(G→A)respectively. There is a SNP locus in exon 8 and it is located on 1251 locus (T→C), which belongs to silent mutation. Two polymorphic loci produce three aggregate genotypes of AA/TT, AB/TC and BB/CC. The correlation analysis reveals that there are significant additive effects on C12:0,C14:0, C16 : 0, C16 : 1, C17: 0, C18 : 2, C20 : 1, C20 :3, PUFA and EFA content between A/T allele and B/C allele. There is the significant additive effect between different UFA. In conclusion, the fatty acid composition of A/T allele is more beneficial to human health compared with B/C allele.
