摘要
目的:评价血尿安胶囊对急慢性肾炎型血尿的防治作用及机制,为临床应用提供实验依据。方法:采用脂多糖(LPS)致小鼠急性肾功能损伤模型、牛血清白蛋白(BSA)加LPS以及四氯化碳(CCl4)等联合诱导血尿大鼠模型考察药物对肾脏相关功能的影响。结果:小鼠注射LPS后,其肾指数、血清BUN及CREA水平显著升高;血尿安胶囊3.0g/kg可显著降低LPS小鼠血清BUN水平。大鼠在联合给予BSA、CCl4及LPS 8周后,其尿蛋白及隐血显著升高,血清CREA水平显著升高,肌酐清除率显著降低,肾指数显著增加,其肾脏IL-1α、IL-1β、IL-10以及MCP表达均显著降低;血尿安胶囊各剂量组可显著降低造模大鼠尿隐血,血尿安胶囊2.5g/kg及5.0g/kg还可显著减少其尿蛋白浓度;血尿安胶囊2.5g/kg可显著降低造模大鼠血清CREA水平,升高其尿CREA水平;血尿安胶囊5.0g/kg可显著降低造模大鼠的尿量及其血清BUN、CREA水平,升高其尿CREA水平;血尿安胶囊10.0g/kg能显著降低造模大鼠肾脏IL-1α、MIP-1α、IL-1β、IL-6、IL-10、IFN-γ、MCP-1、IP-10含量,而血尿安胶囊2.5g/kg能显著升高大鼠肾脏IL-1β、IL-10含量。结论:血尿安胶囊对LPS所致的小鼠急性肾损伤有一定的保护作用,对LPS+BSA+CCl4等联合诱导的大鼠免疫异常性血尿有较好的防治作用,其作用机理可能与对炎症细胞因子的调节有关。
Objective: To investigate the protective effect and its mechanisms of Xueniao'an capsule on hematuria. Methods: ①Acute renal injury was induced by injection of lipopolysaccharide( LPS,10 mg / kg) via tail vein to observe the protective action of Xueniao'an capsule in mouse.② To induce the chronic hematuria in rat,bovine serum albumin( BSA,400 mg /kg) was given by gavage once every other day for 8w,plus CCl4( cooking oil 0. 5ml + CCl40. 1ml) was given by subcutaneous injection once a week for 8w. Meanwhile,LPS( 0. 05 mg) was injected into rat's tail vein at the 5th and the 7th week,respectively. Results: 1 Renal index,serum BUN concentration and serum CREA concentration were increased significantly in LPS-induced acute renal injury mice( P〈0. 01,P〈0. 05). Xueniao'an capsule( 3g / kg) reduced serum BUN concentration in LPS-treated mice( P〈0. 05). 2 Urine protein,occult blood,renal index and serum CREA concentration were increased significantly( P〈0. 01,P〈0. 05),however,creatinine clearance rate and renal cytokines such as IL-1α,IL-1β,IL-10,MCP were decreased significantly( P〈0. 01,P〈0. 05) in BSA + CCl4+ LPS-induced hematuria model rats. Xueniao'an capsule( 10 g / kg,5g / kg,2.5g / kg) reduced occult blood of urine and urine protein in chronic hematuria rats( P〈0. 01,P〈0. 05). Xueniao'an capsule( 5g / kg,2. 5g /kg) also reduced serum BUN and CREA concentration( P〈0. 01,P〈0. 05),but increased urine CREA concentration( P〈0. 01,P 〈0.05) in chronic hematuria rats. Finally,Xueniao'an capsule( 10 g / kg) decreased levels of cytokines such as IL-1α,IL-1β,IL-6,IL-10,IFN-γ,MCP-1,MIP-1α and IP-10( P〈0. 01),however,Xueniao'an capsule( 2. 5g / kg) increased the levels of IL-1β and IL-10( P〈 0.05) in kidney in BSA + CCl4+ LPS-induced hematuria rats. Conclusion: The protection of Xueniao'an capsule against hematuria caused by abnormal immunity may be related to regulating the levels
