摘要
目的:研究桂枝茯苓胶囊及活性成分组合物(15个成分)对子宫肌瘤细胞抑制作用及其机制。方法:对子宫肌瘤细胞进行原代、传代培养及细胞鉴定。CCK-8法分析桂枝茯苓胶囊及活性成分组合物对人子宫肌瘤细胞抑制作用;检测细胞凋亡、线粒体膜电位、caspase-3活性,采用Western Blot法检测Bcl-2蛋白表达情况。结果:桂枝茯苓胶囊(212μg/ml)及活性成分组合物(48、24μg/ml)对子宫肌瘤细胞增殖有抑制作用。桂枝茯苓胶囊及组合物可诱导人子宫肌瘤细胞凋亡、引起人子宫肌瘤细胞线粒体膜电位的下降、子宫肌瘤细胞中Caspase-3活性明显升高,诱导人子宫肌瘤细胞中Bcl-2下调、Bax上调,Bcl-2/Bax比例下降。结论:桂枝茯苓胶囊及活性成分组合物对子宫肌瘤细胞有明显的抑制作用,其作用机制与诱导人子宫肌瘤细胞凋亡有关,该凋亡诱导作用与线粒体信号通路(膜电位下降、Bcl-2/Bax比率降低、Caspase-3活性增强)有关。
Objective: To study the inhibition effects and mechanism of the guizhi fuling capsule and its active ingredient composition( 15 ingredients) on uterine leiomyoma cells. Methods: Human uterine leiomyoma cells were primary cultured subcultured and identified. Viability of cells were determined by CCK-8 to test effects of guizhi fuling capsule and its active ingredient composition on uterine leiomyoma cells. Cell apoptosis,mitochondrial membrane potential,caspase-3 activity were detected. Using Western Blot method to detect the Bcl-2 protein expression. Result: Guizhi fuling capsule( 212,106 μg / ml) and its active ingredient composition( 48,24 μg / ml) could inhibit proliferation of uterine leiomyoma cells and induce uterine leiomyoma cells apoptosis and decline mitochondrial membrane potential and rise Caspase 3 activity( P〈0. 05,P〈0. 01). Guizhi fuling capsule and its active ingredient composition could induce the Bcl-2cut,Bax raised and Bcl-2 / Bax decline( P〈0. 05,P〈0. 01). Conclusion: Guizhi fuling capsule and its active ingredient composition could inhibit proliferation of uterine leiomyoma cells. The mechanism was related to inducing apoptosis in human uterine leiomyoma cells and the induction of apoptosis was associated with mitochondria signal pathway( membrane potential decreased,Bcl-2 / Bax ratio decreased,the activity of Caspase-3 enhanced).
