摘要
目的探讨WT1和eIF4E基因在儿童急性B淋巴细胞白血病中表达情况及意义。方法 73例儿童急性淋巴细胞白血病患者为白血病组,其中初诊患儿42例,完全缓解患儿23例,复发患儿8例。选择同期体检健康儿童20例为对照组,2组采用实时定量RT-PCR法检测WT1和eIF4E基因表达水平,并进行比较。结果白血病组WT1和eIF4EmRNA表达水平[208.9(48.9,3 902.5)、49.0(24.0,493.2)]明显高于对照组[33.7(14.0,99.7)、10.8(3.0,39.4)](P<0.05);初诊和复发患儿WT1基因表达水平[195.8(104.3,988.9)、1 649.9(687.4,4 502.5)]明显高于完全缓解患儿[90.5(44.9,317.0)](P<0.05),复发患儿明显高于初诊患儿(P<0.01);初诊患儿eIF4E基因表达水平[137.8(33.8,673.2)]明显高于完全缓解患儿[27.5(19.0,151.6)]和复发患儿[51.9(24.2,373.2)](P<0.01)。结论WT1基因参与了儿童急性B淋巴细胞白血病的初次发病和复发,eIF4E基因可能仅与初次发病有关。
Objective To explore the expressions of WT1 and eIF4E genes in children with acute lymphocytic leukemia and their clinical significances. Methods Seventy-three children with acute lymphocytic leukemia (leukemia group) included 42 newly diagnosed children, 23 complete remission and 8 recurrent children. Another 20 healthy children were selected as controls (control group). The expression levels of WT1 and eIF4E genes were detected by real time quantitative PCR technique and compared between two groups. Results The levels of WT1 mRNA and eIF4E mRNA were 208.9 (48.93, 902.5)and 49.0 (24.0, 493.2) in leukemia group, significantly higher than those in control group ((33.7 (14.0, 99.7), 10.8 (3. 0, 39.4) (P^0.05). The level of WT1 gene was 195. 8 (104. 3, 988. 9) in newly diagnosed children and 1 649. 9 (687. 4, 4 502. 5) in recurrent children, significantly higher than that in complete remission children (90.5 (44.9, 317.0)) (P〈0.01), and was higher in recurrent children than that in newly diagnosed children (P%0.01). The level of eIF4E gene was significantly higher in newly diagnosed children (137.8 (33.8, 673.2)) than that in complete remission children (27.5 (19.0, 151.6)) and recurrent children (51.9(24.2, 373.2)) (P〈0.01). Conclusion WT1 gene participates in the pathogenesis and recurrence, and eIF4E gene may participate in only the recurrence of acute lymphocytic leukemia in children.
